Characterization and cloning of operon encoding 2, 4- Diacety phlorouglosinol from Pseudomona fluorescens UTP-F 68 Genom

Abstract:
In recent years، certain strains of Florescent Pseudomonas resident in rhizosphere have considerably attracted lots of attention due to their protection role against various types of plant pathogens. The ability of antibacterial metabolites production is one of the important characteristics of these bacteria that plays a key role in plant production against pathogens. Fluorescent pseudomonades are able to produce antibiotics playing a role in the biological control of plant pathogens. 2،4 – diacetylflourogolocinol (DAPG)، is one of the most significant antibiotics controlling soil borne plant diseases which is produced by strains of Pseudomonas fluorescent. The Production of the antibiotic is controlled by six genes in three transcription units. In this study، four genes: phlA، phlC، phlB، and phlD that have direct role in the synthesis of the antibiotic and have been transcribed in one unit and organized in one operon were isolated from Pseudomonas fluorescens genome using PCR. Eventually، Carrying out the validation of the isolation using the Nested PCR technique، the operon was cloned in the pGEMT-easy vector system II for the future studies.
