The effects of anticoagulants on hematological indices of (Huso huso Linnaeus 1758)

Abstract:
Hematological parameters and osmotic fragility in Huso huso blood collected on three anticoagulants, heparin (10 IU/mL), Na2EDTA (0.1, 0.5, and 1 mg/mL), and sodium citrate (0.3 mg/mL), were compared. For this purpose, blood sampling were done by cutting tail fin of fish, without using anesthesia. The highest numbers of WBC were found in Na2EDTA (1 mg/ml). WBC differential count revealed significant difference in monocytes, lymphocytes and eosinophiles (p≤0.05). The differences in hemoglobin blood levels werent significant in treatments of Na2EDTA (p≥0.05). RBC in Na2EDTA (0.1, 0.5 mg/mL) was significantly higher than others (p≤0.05). MCH (pg), MCV (fl) and MCHC (%) in all concentration of Na2EDTA were higher than heparin and citrate. Resistance to hemolysis in heparin- treated was higher than the other. The obtained data suggested heparin as the most appropriate anticoagulant for this species that should be considered in hematological studies.
