The Effect of Conditioned Media from Bone Marrow-Derived Stem Cells in Treatment of Azoospermic infertility Induced by Busulfan in Bulb- C Mice

Background and Aim
Administration of chemotherapy in cancer patients can cause infertility. This study was conducted to evaluate the effect of conditioned media from bone marrow-derived stem cells (secreta) in treatment of azoospermic infertility induced by Busulfan in Bulb–C mice.
Methods
In this experimental study,eighteen Balb-C mice were randomly divided into three groups of control,azoospermia and secreta. The control group did not receive any treatment. In order to stop endogenous spermatogenesis and induce azoospermia, two doses of 10 mg/kg of busulfan with 21 days interval were injected. In secreta group, 35 days after the last injection of busulfan, conditioned media from bone marrow-derived stem cells was injected into the testes. After 8 weeks, testes were removed for histological and histomrphometric evaluations. the collected data were statistically analyzed using one-way analysis of variance.
Results
Histological findings showed, in azoospermia group, spermatogenic cells had dramatically decreased and vacuolated spaces in the germinal epithelium were observed. After treatment with secreta, the germinal epithelium layer and spermatogenesis process significantly improved. The histomrphometric findings indicated that cellular diameter, cellular area, total diameter, cross-sectional area and spermatogenesis index in the seminiferous tubules in the conditioned media were more than azoospermia group (p0.05).
Conclusion
Injected conditioned media from bone marrow-derived stem cells (secreta) into testis could induce spermatogenesis in seminiferous tubules of azoospermic mice. That these findings can be added to the previous literature for treatment of male infertility.
