Effect of Tobacco streak virus on podding of different soybean cultivars under greenhouse conditions

Tobacco streak virus is one of the most important destructive viruses of crops, and reported to be as a causal agent of soybean bud blight worldwide. In this study, the response of common soybean cultivars to TSV and its possible effect on soybean podding under greenhouse conditions was evaluated in the north of the country.This research was carried out in two way factorial arrangement on a CRD (completely randomized design) with virus isolates (soybean, mungbean and sunflower isolates) and soybean cultivars (Williams, Amir, Katol, Saman, Sari) as treatments in 3 replications. Using with the DAS-ELISA method and polyclonal antibody specific for TSVand polymerase chain reaction (PCR) and designed primers specific for virus, plants were tested for TSV infection. Results showed that viral isolates significantly reduced the plant length, number of nodes, number of healthy pods and increased disorder pods. In addition to the growth indices (plant length, number of nodes, pod states, dry weight and fresh weight), viral infection had a significant effect on the numbers of disorder pods in different cultivars.The lowest differences in growth indices and podding disorder observed in Williams cultivar whereas most podding disorder seen in Amir and Katol cultivars. The symptoms caused by soybean virus isolates were more severe than mungbean and sunflower isolates of the virus. Results indicated to the significant effect of viral isolates and soybean cultivars on podding disorder.
