Inhibitory effect of (−)-epigallocatechin-3-gallate (EGCG) on toxicity induced by bisphenol A in bone marrow mesenchymal stem cell in adult rat

Abstract:
Background and aim

Bisphenol A (BPA), known as a xenoestrogen, is widely used in industry and dentistry. Epigallocatechin-3-gallate (EGCG) is a strong antioxidant playing important role in scavenging of free radicals .This study aims to investigate the protective role of EGCG on BPA-induced oxidative stress in mesenchymal stem cells (MSCs).

Methods

After isolation and culture of MSCs from adult rats, cells divided into 4 groups: control, BPA, EGCG, and BPA+EGCG. Then cells were treated for 24, 36, 48, 60, and 72 h and viability, cell morphology and calcium content of cells were evaluated.

Results

BPA caused significant dose- and time-dependent reduction of viability and calcium content. In addition, morphological changes such as cytoplasm shrinkage were observed in the group treated by BPA. EGCG inhibited the cytotoxicity and morphogenic changes induced by BPA.

Conclusion

BPA reduces the viability of mesenchymal stem cells and the antioxidant EGCG reduces the BPA-induced toxic effects.
