The Efficacy Of Clarithromycin against Murine Toxoplasmosis

The effects of clarithromycin alone and in combination with sulfadiazine and pyrimethamine were evaluated against acutemurine Toxoplasmosis.Four groups of mice (each group 20 mice) were separately inoculated intra peritoneally with lX104 tachyzoites of Toxoplasma gondii, RH strain and after 24 hours infection were treated as follows:Group 1: Pyrimethamine 15 mg/kg/day plus sulfadiazine 80 mg/L of drinking water for 10 days.Group 2: Clarithromycin 300 mg/kg/day plus Pyrimethamine 15mg/kg/day for 10 days.Group 3: Clarithromycin 200 mg/kg/day plus sulfadiazine 80 mg/L of drinking water for 10 days.Group 4: Clarithromycin 300 mg/kg/day for 10 days.For each group of mice a control group was selected and inoculated by the parasite but not treated. The results showed that all untreated control mice died within 5 to 9 days after inoculation and Toxoplasma gondii were found in their peritoneal exudate. But 100% of the group 1, 80% of group 2, 70% of group 3 and 30% of group 4 were survived after 5 weeks of infection and no parasite were found in the smears prepared from their viceral organs and their brains.
