صفحات 1-8هدفبررسی تاثیر اجزای پروتئینی و اسید نوکلئیکی توکسوپلاسما گوندی بر بلوغ سلول های دندریتیک و توانایی آن ها در تولید IL-12 و تکثیر لنفوسیت های T اختصاصی آنتی ژن های توموریمواد و روش هاسلول های مغز استخوان موش به مدت پنج روز در حضور GM-CSF و IL-4 کشت داده شدند. روز پنجم سلول های دندریتیک نابالغ به دست آمده با لیزات تومور و اجزای پروتئینی و اسید نوکلئیکی توکسوپلاسما گوندی و لیپوپلی ساکارید به مدت دو روز کشت داده شدند. برای بررسی عملکرد سلول های دندریتیک در تحریک لنفوسیت های T از آزمون MLR استفاده شد. از کیت الایزا برای اندازه گیری میزان IL-12 استفاده و بلوغ سلول های دندریتیک با فلوسیتومتری بررسی شد.نتایجسلول های دندریتیک بالغ شده با اجزای پروتئینی توکسوپلاسما باعث تکثیر معنی داری در سلول های طحالی در مقایسه با دیگر گروه ها شد و میزان تولید IL-12 در این گروه بالاتر بود (001/0>P).نتیجه گیریترکیبات مختلف پیکره میکروبی مانند اجزای پروتئینی و اسید نوکلئیکی توکسوپلاسما گوندی می توانند باعث افزایش قابلیت عرضه آنتی ژن سلول های دندریتیک به لنفوسیت ها شوند که در این میان تاثیر اجزای پروتئینی از اجزای اسید نوکلئیکی بیشتر بوده است.

صفحات 9-16هدفتب راجعه کنه ای بیماری حاد عفونی است که توسط اسپیروکتی به نام بورلیا پرسیکا ایجاد و توسط کنه های نرم اورنیتودوروس تولوزانی منتقل می شود. این بیماری در خاورمیانه و نیز مناطق زیادی از کشور ایران دیده می شود. یکی از مشکلات موجود درباره این بیماری تعیین آلودگی در کنه ها برای تعیین ناقل بیماری است که به روش کلاسیک گزنودیاگنوزیز انجام می شود که عبارت است از خون دهی کنه ها روی حیوان حساس آزمایشگاهی و بررسی میکروسکوپی خون حیوان پس از 7-14 روز، که روشی ناکارامد و طولانی مدت است. در این مطالعه کارایی دو روش مولکولی PCR و کلاسیک گزنودیاگنوزیز برای تعیین آلودگی کنه های اورنیتودوروس تولوزانی به بورلیا پرسیکا مقایسه شده است.مواد و روش هانمونه های کنه از مناطق شمال غرب کشور جمع آوری و روی خوکچه هندی آلوده به بورلیا خونخواری نمودند. برای آزمایش های کلاسیک پس از هضم کامل خون، این کنه ها مجددا روی خوکچه های سالم خونخواری نموده و پس از 5 تا 14 روز، آلودگی خون آن ها به بورلیا با بررسی های میکروسکوپی مشخص شد. برای PCR، DNA بورلیا همراه با DNA کنه ها استخراج و به کمک آغازگرهای اختصاصی ژن 16S-rDNA، ژنوم بورلیا تکثیر و سپس تعیین توالی شد. در این مطالعه همچنین تاثیر جنس کنه (ماده و نر)، گذشت زمان و حداقل آلودگی لازم برای عملکرد PCR ارزیابی شد.نتایجبررسی های مولکولی به کمک آغازگرهای اختصاصی ژن 16S-rDNA نشان داد که روش PCR می تواند آلودگی را در تمامی کنه های آلوده مشخص نماید در حالی که روش کلاسیک قادر به تشخیص آلودگی در (3/13 درصد) نمونه های آلوده بود. جنس کنه (ماده و نر) و گذشت زمان پس از خونخواری (بلافاصله تا هضم کامل خون) تاثیری بر نتایج واکنش PCR نداشت. ضمن این که وجود یک بورلیا کافی بود تا نتیجه واکنش PCR مثبت شود.نتیجه گیریاین مطالعه اولین ارزیابی تشخیص مولکولی عامل تب راجعه بومی به روش PCR در ایران است و به لحاظ سرعت، دقت و کارایی بالا می تواند جایگزین روش کلاسیک شود و برای تشخیص آلودگی در ایران و سایر مناطق آلوده در خاورمیانه توصیه شود.

صفحات 17-31هدفاین مطالعه به منظور بررسی امکان تشخیص نشانگان داون پیش از تولد با روش Real-Time PCR انجام شد. در این راستا، تعیین روش مناسب برای تخلیص DNA از نمونه های آمنیوسیت برای دستیابی به DNA با کیفیت بالا نیز مورد توجه قرار گرفت.مواد و روش هاابتدا زنان بارداری که در غربالگری از طریق سنجش نشانگرهای بیوشیمیایی و سونوگرافی خطر بالای ابتلای جنین به نشانگان داون را داشتند، به مرکز منیوسنتز معرفی شده و از مایع آمنیوتیک آن ها نمونه گیری شد. تخلیص DNA از 59 نمونه مایع آمنیوتیک با روش های مختلف انجام شد که این روش ها شامل روش جوشاندن، روش رسوب دهی با نمک، دستورالعمل استخراج DNA از خون و بافت با کیت DNPTM (سیناژن)، دستورالعمل استخراج DNA از سلول با کیت DNA Isolation Kit for cells and tissues (Roche)، دستورالعمل استخراج DNA از بافت با کیت MagNa Pure DNA Isolation (Roche) و کیت QIAamp DNA Micro (Qiagen) بودند. سپس کیفیت و کمیت نمونه های تخلیص شده به وسیله دستگاه اسپکتروفتومتر نانودراپ NanoDrop® ND-1000 ارزیابی شد. واکنش Real-Time PCR با استفاده از رنگ سایبرگرین I (Applied Biosystems، UK) به منظور تکثیر اختصاصی ژن های DYRK1A2 و DSCAM و ژن مرجع PMP22 روی نمونه های تخلیص شده انجام شد. آنالیز داده ها با استفاده از روش مقایسه ای چرخه آستانه برای تعیین میزان ژنی و تعیین تعداد نسخه های کروموزوم 21 صورت گرفت.نتایجاین بررسی نشان داد که DNA استخراج شده از نمونه های مایع آمنیوتیک با استفاده از کیت QIAamp DNA Micro دارای کمیت و کیفیت مطلوب است. تکثیر اختصاصی ژن های هدف و مرجع انجام و تفاوت گروه طبیعی و گروه مبتلا براساس اختلاف در چرخه آستانه مشخص شد.نتیجه گیریتشخیص پیش از تولد با روش Real-Time PCR روی نمونه های DNA تخلیص شده از سلول های مایع آمنیوتیک با استفاده از کیت Qiagen امکان پذیر است. نتایج حاصل با نتایج مشابه حاصل از روش های متداول سیتوژنتیک قابل مقایسه است.

صفحات 33-39هدفروش تداخل RNA یا RNAi بهترین روش خاموش کردن ژن ها در سطح ترانس کریپتومی شناخته شده است، سرطان ها و بیماری های ژنتیکی اهداف مهمی برای توسعه روش های درمانی مبتنی بر RNAi به شمار می روند. برای حل مشکل خاموشی موقتی سیستم های siRNA، RNA های سنجاق سری یا shRNA ها ابداع شدند و نسل جدید shRNA ها به نام shRNAmir است. سازه خاموش گر با ساختار سنجاق سری شبه microRNA (shRNAmir)، رفتاری مشابه یک microRNA طبیعی را درون سلول تقلید می کند. کمک فعال کننده گیرنده استروئیدی (SRA) یکی از تنظیم کننده های گیرنده استروئیدی از جمله گیرنده استروژنی (ER) است. بافت های پروستات، رحم و سینه بیان پایینی از SRA دارند، در حالی که طی تومورزایی آن ها بیان SRA افزایش می یابد، بنابراین امکان شرکت SRA در تومورزایی یا پیشروی توموری وجود دارد.مواد و روش هادر مطالعه حاضر از روش RNA تداخل گر یا RNAi برای خاموش نمودن ژن SRA استفاده شد و سازه خاموش گر SRA طراحی و توسط Soe-PCR ساخته شد و سپس در پلاسمید pEGFPC1 که یک ناقل بیانی است کلون گردید. رده سلولی انسانی سرطان پستان (MCF7) با پلاسمید حاصل ترانسفکت شد و بیان SRA در زمان های 24، 72 ساعت و 10روز بعد با Real-Time PCR بررسی شد.نتایجکاهش 60 درصدی در بیان نسبی SRA در تیمارهای 72 ساعت و 10روز بعد مشاهده شد که نشان می دهد shRNAmir-SRA با موفقیت توانسته است بیان ژن هدف مطالعه حاضر را به طور نسبتا پایداری خاموش کند.نتیجه گیریبا توجه به موفقیت در طراحی و ساخت یک سازه shRNAmir و خاموشی نسبتا پایدار ژن هدف، این سازه برای مطالعات مختلفی در آینده آماده و مناسب به نظر می رسد.

صفحات 41-49هدفبا توجه به آثار مرگ بار آفلاتوکسین ها و به خصوص آفلاتوکسین B1 بر سلامتی انسان، اندازه گیری آفلاتوکسین متصل به آلبومین، به عنوان یک شاخص مهم میزان در معرض بودن به آفلاتوکسین ضروری به نظر می رسد. هدف از این پژوهش، بهینه سازی روش کروماتوگرافی مایع با کارایی بالا و آشکارگر فلورسانس به منظور اندازه گیری این شاخص در خون است.مواد وروش هادر این مطالعه، از نمونه سرم خون سه گروه رت به عنوان کنترل مثبت (تیمار شده با آفلاتوکسین B1)، کنترل (بدون تیمار خاص) و استاندارد (تیمار شده با میزان معینی آفلاتوکسین B1 نشان دار با تریتیوم) استفاده شد. پس از جداسازی آلبومین از نمونه سرم با استفاده از آمونیوم سولفات و اسید استیک، خلوص آلبومین با الکتروفورز SDS-PAGE بررسی و غلظت آن به روش برادفورد اندازه گیری شد. سپس آلبومین تحت تاثیر پروناز هیدرولیز شد تا آفلاتوکسین متصل به آلبومین به صورت آفلاتوکسین- لیزین آزاد شود. سپس آنزیم پروناز و باقیمانده حاصل از هیدرولیز آلبومین توسط استون در سرما رسوب داده و جدا شد. حجم محلول رویی را با دستگاه فریز- درایر تقلیل داده و سپس نمونه تغلیظ شده به دستگاه HPLC تزریق و مقدار آفلاتوکسین موجود در آن در مقایسه با نمونه مشابه حاصل از رت های گروه استاندارد، اندازه گیری شد.نتایجبا الکتروفورز SDS-PAGE، خلوص آلبومین جدا شده اثبات شد و غلظت آن در نمونه های کنترل مثبت، منفی و استاندارد به ترتیب 10، 5/12 و 13 میلی گرم در میلی لیتر به دست آمد. در روش HPLC حداقل میزان تشخیص (Detection limit) برای اندازه گیری آفلاتوکسین متصل به آلبومین، 20 پیکوگرم در هر میلی گرم آلبومین، اختصاصیت روش، 92 درصد و حساسیت آن، 100 درصد تعیین شد. میانگین میزان آفلاتوکسین متصل به آلبومین در سرم رت های کنترل مثبت، 10 نانوگرم در هر میلی گرم آلبومین محاسبه شد و تکرارپذیری روش طی بارها تکرار، بسیار خوب ارزیابی شد.نتیجه گیریدر این تحقیق برای اندازه گیری آفلاتوکسین متصل به آلبومین به روش HPLC تغییراتی در فاز متحرک، درصد حلال ها و زمان هر آزمایش ایجاد شد و همچنین کروماتوگرافی میل ترکیبی قبل از انجام HPLC حذف شد. بر این اساس HPLC-Fluorescence به عنوان یک روش دقیق، حساس و اختصاصی و با بهینه سازی ایجاد شده با سرعت و سهولت بیشتر، تکرارپذیری بالاتر و هزینه کمتر می تواند برای اندازه گیری آفلاتوکسین B1 در سرم استفاده شود.

صفحات 51-60هدفمهندسی بافت، علمی میان رشته ای است که اساس آن مبتنی بر به کارگیری داربست های پلیمری برای شکل گیری سه بعدی بافت است. از این داربست ها به عنوان ماتریکس خارج سلولی سنتتیک برای چسیندگی سلول ها، تکثیر و تمایز سلولی استفاده می شود. مواد زیستی مورد استفاده در مهندسی بافت می بایست از ترکیب شیمیایی، ساختار فیزیکی و عملکرد زیستی مناسب برخوردار باشند. در پروژه حاضر، ترکیب کیتوزان/پلی وینیل الکل به عنوان داربست برای ترمیم بافت عصبی انتخاب شده است.مواد و روش هابرای تولید داربست های کیتوزان/پلی وینیل الکل با استحکام مکانیکی و ریخت شناسی مناسب در کاشت و تکثیر سلول های عصبی U373 از روش الکتروریسی استفاده شد. روش الکتروریسی نسبتا ساده بوده و می توان به وسیله آن الیافی به فرم لایه نازک بی بافت تولید کرد. پس از آن میزان زیست سازگاری داربست با استفاده از آزمون های زیستی و ارزیابی میزان چسبندگی سلولی مورد بررسی قرار گرفت.نتایجنتایج کسب شده نمایان گر آن هستند که نانوکامپوزیت کیتوزان/پلی وینیل الکل با نسبت 85/15 ضمن تامین ریخت شناسی و استحکام مکانیکی لازم، امکان رشد سلول های عصبی U373 به فضای درونی ماده زیستی و جایگزینی آن ها را در طول زمان و به صورت کنترل شده فراهم می کند. بدین ترتیب شرایط لازم برای رشد و توسعه سلولی و همچنین جدا شدن از بافت رشد یافته فراهم می شود.نتیجه گیریبا استفاده از نانوکامپوزیت کیتوزان/پلی وینیل الکل به دلیل زیست سازگاری مناسب و عدم سمیت، امکان رشد سلول های U373 و اتصال مناسب آن ها به نانوکامپوزیت برای ترمیم آسیب وارده به اعصاب محیطی فراهم می شود.

صفحات 61-69هدفاستافیلوکوکوس اورئوس از عوامل مهم عفونت های شدید در بیمارستان و جامعه است. سویه های مقاوم به متی سیلین این باکتری شیوع و مرگ و میر بالا دارند. بنابراین برای درمان آنتی بیوتیکی و کنترل پخش عفونت لازم است که سویه های مقاوم به متی سیلین به سرعت و دقت شناسایی شوند. در این تحقیق مقاومت به متی سیلین با روش انتشار دیسک، آگار دایلوشن و PCR برای ژن mecA بررسی شد.مواد و روش ها174 سویه استافیلوکوکوس اورئوس از نمونه های مختلف بالینی از سه بیمارستان آموزشی جدا شد. حساسیت آنتی بیوتیکی با روش انتشار دیسک تعیین شد و حداقل غلظت مهارکنندگی اگزاسیلین با آگار دایلوشن و PCR برای ژن mecA با آغازگرهای اختصاصی انجام شد.نتایجفراوانی سویه های مقاوم به متی سیلین با روش انتشار دیسک، آگار دایلوشن و PCR به ترتیب 50 درصد، 7/47 درصد و 2/48 درصد مشاهده شد و سویه های mec مثبت نسبت به آنتی بیوتیک های آزمایش شده بسیار مقاوم تر از سویه های mec منفی بودند. نتایج آگار دایلوشن، مقاومت سطح پایین به متی سیلین (حداقل غلظت مهارکنندگی کمتر از 64 میلی گرم در لیتر) را نشان داد. پراکندگی سویه های مقاوم به متی سیلین در سه بیمارستان مورد بررسی مشابه بود و اختلاف معنی دار بین حضور سویه های مقاوم به متی سیلین در نمونه های مختلف بالینی مشاهده نشد.نتیجه گیریPCR بهترین روش برای شناسایی روتین سویه های مقاوم به متی سیلین است به طوری که می تواند 24 ساعت قبل از سایر روش های معمول سویه های مقاوم به متی سیلین را شناسایی کند.

صفحات 71-77هدفدر تحقیق حاضر اثر بازدارندگی از رشد پلی فنل های برگ سبز چای به عنوان مواد ضد قارچی گیاهی بر مخمر فرصت طلب کاندیدا آلبیکنس (سویه استاندارد PTCC-5027) در دو زمان 24و 48 ساعت بررسی شد.مواد و روش هابا استفاده از آزمون حساسیت دارویی با روش ماکرودیلوشن براث (رقت در لوله)، حداقل غلظت کشندگی ضد قارچی (MFC) و کمترین غلظت مهارکنندگی از 90 درصد رشد قارچ (MIC90) برای پلی فنل های برگ سبز چای و فلوکونازول علیه کاندیدا آلبیکنس (PTCC-5027) در دو زمان 24 و 48 ساعت بررسی شد.نتایجفعالیت ضد قارچی کاتشین (مؤثرترین ترکیب برگ سبز چای) وابسته به زمان است. کمترین غلظت مهارکنندگی کاتشین در مقادیر 103×5/0، 103×1 و 103×2 مخمر در میلی لیتر پس از 24 ساعت به ترتیب 5/12، 25 و 100 میلی لیتر در میلی گرم و پس از 48 ساعت به ترتیب 25/6، 5/12 و 50 میلی لیتر در میلی گرم به دست آمد. مخمر نسبت به فلوکونازول مقاوم گزارش شد. کلیه نتایج با محاسبات میانگین آماری، پس از 3 بار تکرار ثبت شد.نتیجه گیریبا وجود مقاومت استرین کاندیدا آلبیکنس مورد مطالعه نسبت به فلوکونازول، پلی فنل های برگ سبز چای خصوصا کاتشین، به خوبی از رشد این مخمر ممانعت کرده و کاهش رشد در غلظت های MIC90 و MFC مشهود است. براساس نتایج به دست آمده، برگ سبز چای حاوی ترکیبات مؤثر ضد قارچی بوده و از آن جایی که داروهای ضد قارچی رایج، دارای آثار سوء جانبی هستند و از طرفی نیز شاهد افزایش مقاومت های دارویی هستیم، امید است با ساخت داروهای گیاهی از مواد مؤثر آن ها، جایگزین خوبی برای درمان بیماری های قارچی تجویز شود.

Pages 1-8ObjectiveThe aim of the present study was to investigate the effect of protein and DNA components of Toxoplasma gondii on maturation of dendritic cells and their efficiency in IL-12 production and proliferation of T cells.Materials And Methodsfor DC generation, Bone marrow cells were cultured in the presence of GM-CSF and IL-4 for 5 days. Tumor lysate and protein or DNA components of Toxoplasma gondii were added to the culture media and incubated for another 2 days. LPS was added as control for DC maturation. Proliferation of T cells were determined by MLR and IL-12 production was measured by ELISA kit. Maturation of dendritic cell were determined by flowcytometry.ResultsDCs treatment with protein components of Toxoplasma gondii caused a significant increase in IL-12 production and proliferation of T cells (P<0.001).ConclusionsDifferent compositions of microbial body like protein and DNA components of Toxoplasma gondii can cause augmentation of antigen presentation capacity of DC and their IL-12 production capability. Among these components the protein was more effective as compared to DNA.

Pages 9-16ObjectiveRelapsing fever caused by Borrelia persica is an acute tick-borne disease which is transmitted by soft ticks of Ornithodoros tholozani to human. The disease is reported from Middle East and many regions of Iran. Detection of infection is problematic since the suspected infected ticks should be fed on animal hosts such as guinea pigs and subsequently after 7-14 days, the animal blood should be microscopically investigated for Borellia spirochetes on a Giemsa stainined thick smear. This classic method named xenodiagnosis is hard, time consuming, and less reliable. In this study, the application of PCR technique has been examined for detection of Borellia persica in soft ticks of O. tholozani.Materials And MethodsTick specimens were collected from northwestern Iran and were fed on Borellia persica infected guinea pigs. DNA of the animal blood were extracted and used as target for PCR amplification of 16rDNA gene. Subsequently the products were subjected to sequencing. The effect of tick sex and post digestion as well as the minimum number of spirochetes on the efficiency of PCR were also tested.ResultsThe xenodiagnosis assay was able to detect infection in only 13.3% of the tick-bitten animal bloods whereas all of these blood specimens were PCR positive against the 16rDNA gene. There wasno difference in results of PCR for male and female of the ticks. Post digestion of infected blood meal in ticks did not affect the efficacy of PCR and the recently-fed samples showed similar results to those of completely gravid ones. A test on the threshold sensitivity of PCR assay indicated that only one spirochete is enough for the primers to anneal and to amplify the target gene.ConclusionThis study describes the first molecular assay for diagnosis of B. persica infected ticks in Iran and due to its high speed, accuracy, and applicability is a substitution method for diagnostic purposes in TBRF foci.

Pages 17-31ObjectiveIn this study, the possibility of prenatal diagnosis of Down syndrome with Real-Time PCR method was evaluated. In this context, optimization of a suitable method for purification of high quality DNA from amniotic fluid samples was also considered.Materials And MethodsPregnant women who had the high risk of having babies with Down syndrome were selected according to the biochemical and sonographic data and referred to the amniocentesis center. The DNA of total 59 amniotic fluid samples were extracted with different methods including boiling method, salting out method, Procedures of DNA extraction from Blood and Cell Culture by DNPTM Kit (CinnaGen), Procedure of DNA extraction from cells by DNA Isolation Kit for cells and tissues (Roche), Procedure of DNA extraction from Tissue by MagNa Pure DNA Isolation kit (Roche), and QIAamp DNA Micro Kit (Qiagen). Then, the quality and quantity of the extracted DNA were evaluated by the NanoDrop® ND- 1000 spectrophotometer device. Real-Time PCR reaction using fluorescent dye SYBR Green I (Applied Biosystems, UK) was performed to specifically amplify DSCAM and DYRK1A2 genes and the reference gene (PMP22). Data analysis was performed using comparative cycle threshold method for the determination of the gene dosage and determining the number of copies of chromosome 21.ResultsThis study showed that DNA extracted from amniotic fluid samples using QIAamp DNA Micro Kit (Qiagen) has the desirable quantity and quality for Real-Time PCR. Specific proliferation of targets and reference genes was achieved and difference between normal and affected groups based on differences between their gene dosages was determined.ConclusionPrenatal diagnosis of Down syndrome is feasible by the Real-Time PCR method using DNA samples from amniotic fluid cells extracted by QIAamp DNA Micro Kit (Qiagen). The results are comparable to the corresponding results from conventional cytogenetic methods.

Pages 33-39ObjectiveRNA interference (RNAi) is the most potent technique for gene silencing in eukaryotic cellular system at transcriptomic level. Genetic disorders and cancers are important targets for therapeutic development of this technique. In order to bypass the temporary dpwnregulation by siRNA, a new generation of shRNA named shRNAmir has developed. Silencing construct with structure similar to microRNA (shRNAmir), mimics a natural microRNA pathway inside the cell. Steroid receptor RNA activator (SRA) is one of the regulators of steroid receptor like ER. Prostate, uterus and breast tissue express a low level of SRA, there is an increase of expression during their tumorgenesis. So SRA may participate in tumorgenesis or proliferation of tumors.Materials And MethodsWe used RNAi technique to silence expression of SRA. The SRA silencer was designed and constructed by Soe-PCR, then cloned into an expression vector pEGFPC1. Human breast cancer (MCF7) cells were transfected with silencer plasmid then the changes in the SRA expression estimated by Real-Time PCR at 24, 72 hours and after 10days.ResultThe results showed about 60% decrease in relative expression of SRA gene, after 72 hours and 10 days, which shows that shRNAmir–SRA could successfully knockdown the expression of target gene.ConclusionIt seems that the designed shRNAmir may be a suitable tool for a variety of applications because it could stably knockdown the expression of target gene.

Pages 41-49ObjectiveWith consideration of lethal effects of aflatoxins specially B1 on human health. Estimation of aflatoxin-albumin adduct, as an important marker of aflatoxin exposure, seems essential. The aim of this study is optimization of HPLC-fluorescence method for measurement of this important marker in blood serum.Materials And MethodsIn this study, blood serum of three groups of rats as A) positive controls (treated with AFB1), B) negative controls (without treatment) and standard rats (treated with radiolabeled AFB1) were used. After albumin isolation using ammunium sulphate and acetic acid, purity of albumin was tested by SDS-PAGE electrophoresis and albumin concentration was quantified by bradford method. Then albumin was hydrolysed by pronase and aflatoxin bound to albumin was released as aflatoxin-lysine. Pronase was precipitated and albumin was digested by aceton in cold, the volume of supernatant was reduced by freeze-drier and injected into HPLC system. Aflatoxin was quantified in comparison to standard rats samples.ResultsThe purity of this isolated albumin was confirmed by SDS-PAGE electrophoresis. Albumin concentration in positive, negative and standard samples were 10, 13 and 12.5 mg/ml, respectively. Detection limit (20 pg/mg Alb) for measurement of aflatoxin was determined by HPLC method, specificity and sensitivity of method were 92% and 100% respectively. The mean concentration of AF-Alb adducts in serum of positive control rats was 10 ng/mg Alb and the reproducibility of the method after several repeat was very good.ConclusionIn this study, for AF-Alb adduct quantification by HPLC method, mobile phase, percentage of solvents and run time were changed and the affinity chromatography before HPLC, was deleted. Therefor HPLC- fluorescence which is a precise and specific method, and since it is fast, highly reproducible and cost effective, also with improvement made, could easily be used for the quantification of this important marker in serum.

The scaffold provides the microenvironment (synthetic temporary extracellular matrix) for regenerative cells, supporting cell attachment, proliferation, differentiation, and neo tissue genesis due to their suitable chemical, physical and biological structures. In this study, chitosan/poly (vinyl alcohol) (CS/PVA) was exploited as scaffold for nerve regeneration.Materials And MethodsElectrospinning was used to fabricate CS/PVA nanocomposites for U373 cells seeding and proliferation. Electrospinning is a versatile and simple method to fabricate non-woven thin layer fibers from polymeric solutions. Consequently, the biocompatibility of CS/PVA nanocomposite was evaluated using biological assays and cell attachment study.ResultsResults indicated that CS/PVA nanocomposites with 15/85 proportion shown an almost homogenous network of the electrospun fibers and confirmed that they can be knitted in meshes and improve U373 cells proliferation and cell attachment.ConclusionThe nano-sized CS/PVA scaffolds are nontoxic and biocompatible which can promote proliferation of U373 cells and their appropriate adhesion to nanocomposite for improved peripheral nerve regeneration.

Pages 61-69ObjectivesStaphylococcus aureus is an important cause of serious infection in both hospital and the community. Methicillin-resistant S. aureus (MRSA) is associated with high morbidity and mortality rates with rapid development of resistance. There is a need for early and reliable detection of MRSA infection to direct antibiotic therapy, and more effectively control cross-infection. In this study, resistance to methicillin was detected by a disk diffusion method, the determination of MIC, and the PCR for mecA gene.Materials And MethodsA total of 174 S.aureus strains were isolated from different clinical specimens from three teaching Hospitals. Antibiotic susceptibility was determined by disk diffusion method, MIC for oxacillin was made by the agar dilution, and mecA gene was identified by specific primers.ResultsThe prevalence of MRSA by three methods ranged from 47% to 50%, and mecA positive isolates were more resistant to all of the antibiotic tested than mecA negative isolates. All S. aureus isolates were resistant to penicillin, and susceptible to vancomycin. The results of agar dilution test indicated a low-level resistance to methicillin (MIC>64mg/l). The distribution of MRSA isolates were uniform between three hospitals, and there were not significant differences in the presence of MRSA between isolates from different clinical specimens.ConclusionThe PCR method was the best test for routine detection of MRSA in the present study. An additional benefit of the mecA PCR is the potential to generate a susceptibility report, 24h earlier than the time of generation of results of conventional susceptibility testing methods.

Pages 71-77ObjectiveIn this study the susceptibility of Candida albicans to inhibitory effect of polyphenols under varying time (24 and 48 hours) conditions were evaluated.Materials And MethodsGreen tea leaf polyphenols were extracted and analyzed by chromatography. Among polyphenols, Catechin showed stronger antifungal activity against C. albicans PTCC-5027. Catechin's MIC90 (The concentration of Catechin causing 90% growth inhibition of tested strain of C. albicans) and MFC (The minimum antifungal susceptibility of Catechin) were determined by Macro dilution test and calculation after 24 and 48 hours.ResultsThe antifungal activity of Catechin was time dependent. Catechin's MIC for 0.5×10³, 1×10³ and 2×10³ cells/ml was 12.5, 25 and 100 mg/ml after 24h respectively. The results after 48h for 0.5×10³, 1×10³ and 2×10³ cells/ml were 6.25, 12.5 and 25 mg/ml respectively. Fluconazol was tested on C. albicans PTCC-5027 and the results indicated that this strain of Candida is fluconazol resistant. Data shown are from three separate experiments and were analysed statistically.ConclusionC. albicans PTCC-5027 is fluconazol resistant, however green tea leaf polyphenols especially Catechin could inhibit the growth of this yeast at MIC and MFC concentrations. On the basis of the obtained results the green tea leaf contains effective antifungal components. Since the common and generic antifungal drugs possess some side effects and also there is an increasing drug resistance, it is hoped that consumption of herbal drugs may help to cure fungal diseases and to avoid the side effects of antimycotics as a good replacement.
