صفحات 17-22مقدمههر چند تاکنون تحقیقات گسترده ای در مورد مارکر های سرطان ریه انجام شده اما هنوز یک مارکر مشخص کلینیکی در این مورد یافت نشده است. هدف از این مطالعه ارزیابی حساسیت و اختصاصیت توام و انفرادی دو بیو مارکرموجود در خون محیطی از جنس mRNA با عناوین آنتی ژن کارسینوآمبریونیک(CEA) و پروتئین X اختصاصی ریه (LUNX) به منظور تشخیص سرطان ریه بوده است.روش کاردر این تحقیق 30 بیمار مبتلا به سرطان ریه و 30 فرد سالم مورد بررسی قرار گرفته اند. بعد از خونگیری و استخراج RNA تام از هر نمونه سه ویال cDNA آماده گردید. ویال های مربوط به هر نمونه با استفاده از تکنیک Real Time RT-PCR مورد آزمایش قرار گرفته و نتایج مربوط به هر ویال از لحاظ حساسیت مارکر مورد نظر مقایسه گردیدند.نتایجوجود mRNA آنتی ژن کارسینو آمبریونیک (CEA) در 24 نفر از 30 فرد مبتلا به سرطان ریه مشاهده شد. بنابراین حساسیت این آنتی ژن 80% تعیین گردید که به طور معنی داری متفاوت از افراد سالم می باشد. mRNA مربوط به این مارکر تنها در 11 نفر از افراد سالم مشاهده شد. حساسیت کلی مربوط به این مارکر به طور معنی داری مرتبط با ویال های شماره دو و سه بود. وجود mRNA مربوط به پروتئین X اختصاصی ریه (LUNX) در 21 نفر از سی نفر بیمار مبتلا به سرطان ریه مثبت بود که نشان دهنده حساسیت 70% این مارکر می باشد و این یافته به طور معنی داری متفاوت از افراد سالم بوده است. حساسیت کلی مربوط به این مارکر به طور معنی داری مرتبط به ویال های شماره 1 و 3 می باشد. در 93.3% از بیماران مطالعه شده حداقل یکی از مارکرها مثبت بود.نتیجه گیریmRNAهای مطالعه شده در این تحقیق که در خون محیطی موجود می باشند به عنوان مارکرهای حساس و اختصاصی به منظور تشخیص اولیه سرطان ریه توصیه می شوند.

صفحات 29-34مقدمهاستفاده از سلول های بنیادی در ضایعات مغزی ناشی از ایسکمی هیپوکسیک در نوزاد انسان می تواند به عنوان یکی از روش های درمانی مورد نظر قرار گیرد.روش کاردر این مطالعه، جهت بررسی اثر درمانی سلول های بنیادی خون بند ناف انسان از موش های صحرایی 14 روزه ای استفاده گردید که به منظور ایجاد مدل ایسکمی هیپوکسیک مغزی نوزادان شریان کاروتید راست حیوانات مورد مطالعه به مدت 1 ساعت مسدود گردید. 7 روز پس از ایجاد ایسکمی، حیوانات در دو گروه مورد تزریق وریدی 105×2 سلول خون بند ناف انسانی و یا نرمال سالین قرار گرفتند. حیوانات در گروه کنترل هیچ ماده ای دریافت نکردند. پس از گذشت دو هفته، با استفاده از دو آزمون حرکتی حیوانات مورد بررسی قرار گرفتند. متعاقبا، حیوانات جهت انجام ارزیابی بافتی و ایمنوهیستوشیمی قربانی شدند.نتایجیافته های ایمنوهیستوشمی بیانگر جایگیری انتخابی سلول های خون بند ناف انسانی تزریق شده در محل ضایعه و کاهش حجم ناحیه ایسکمیک می باشد. 7 روز پس از جراحی، نتایج حاصل از رفتار های حرکتی در گروه تحت درمان (0/3 ± 12/7) در مقایسه با گروه شم (0/05 ± 10) بهبود معنی داری را نشان داد (0/05 < P) و این تفاوت در روز 14در گروه تحت درمان در مقایسه به گروه شم (0/5 ± 11/9) به0/3 ±15/3 رسید. در بررسی عدم تقارن در حرکت و وضعیت ایستای بدن درصد چرخش به راست حیوانات در گروه تحت درمان در مقایسه با گروه شم به ترتیب در روز های 7 (75% در مقایسه با 97%) و 14 (59% در مقایسه با 96%) به صورت معنی داری کاهش یافت (0/05 < P).نتیجه گیرینتایج این مطالعه توانایی سلول های بنیادی مشتق از خون بند ناف انسان را در کاهش ضایعات عصبی مرتبط با ایسکمی هیپوکسیک نوزادان نشان داد.

صفحات 35-44مقدمهسلول های CD4+ T و CD8+ انواع اصلی لنفوسیت ها در ایمنی سلولی می باشند و نقش محوری را در القاء پاسخ های ایمنی کارامد برضد تومور ایفاء می کنند. فراوانی زیر گروه های سلول T در خون محیطی، بافت تومور و غدد لنفاوی زهکشی کننده (dLNs) می توانند بعنوان شاخص های مفید برای ارزیابی سیستم ایمنی در سرطان در نظر گرفته شود.روش هادر این مطالعه، فراوانی سلول های CD4+ T و CD8+ در نمونه های خون، بافت تومور و dLNs بیماران مبتلا به سرطان پستان با یکدیگر مقایسه و با بافت های مشابه از افراد سالم مقایسه شدند. فنوتیپ ایمنی توسط فلوسایتومتری انجام شد و سطوح بیان CXCL10، گرآنزیم B و ماماگلبولین توسط real time PCR ارزیابی شد.نتایجدر خون محیطی، تفاوتی در زیر جمعیت های سلول T بین بیماران و افراد سالم وجود نداشت. فراوانی سلول های CD8+ در بافت تومور نسبت به بافت سالم پستان بالاتر بود، در حالیکه بیان گرآنزیم B مشابه بود. بر مبنای نتایج سطح بیان ماماگلبولین، dLN ها به دو دسته ماکرو- و میکرو-متاستاتیک تقسیم شدند. ما بطور معنی داری فراوانی کمتری از CD4+ در dLN ها ماکرو-متاستاتیک را نسبت به dLN ها میکرو-متاستاتیک یافتیم. فراوانی CD8+ در هر دو dLN مشابه بود، اگرچه بیان گرآنزیم B در dLN ها میکر-متاستاتیک بالاتر بود. تفاوت معنی داری در بیان CXCL10 بین دو نوع dLN وجود نداشت.نتیجه گیریبرمبنای نتایج ما اگرچه تومور بر ایمنی سیستمیک تاثیری ندارد، سلول های توموری سیستم ایمنی موضعی در بافت تومور و dLN ها متاستاتیک را تحت تاثیر قرار می دهند.

صفحات 45-50مقدمهنخود یکی از مهمترین محصولات کشاورزی با ارزش زیاد غذایی است. فقدان اسانس در دانه نخود موجب می شود که در کاوش مولکولهای دارویی دارای سمیت کم این گیاه مورد توجه قرار گیرد. در این مطالعه اثر ضد تشنجی دانه نخود در مدل های تجربی صرع بررسی شده است.روش هااز دانه ها عصاره دی کلرومتانی به روش پرکولاسیون تهیه شد. سمیت حاد عصاره در موش سنجیده شد. اثر ضد تشنجی عصاره بر علیه تشنجات تونیک ناشی از شوک الکتریکی حداکثر (با شدت 50 میلی آمپر، تواتر50 هرتز و مدت تحریک نیم ثانیه)، تشنجات کلونیک ناشی از پنتیلن تترازول (60 میلی گرم بر کیلوگرم، داخل صفاقی) و مدل صرعی کیندلینگ الکتریکی در موشها بررسی شد. سپس با ان-هگزان از عصاره دو فراکشن f1 و f2 استخراج شد. اثر ضد تشنجی فراکشنها در سه مدل تشنجی ارزیابی شد. آزمایشهای فیتوشیمی بر روی عصاره و فراکشنها انجام شد. ساختار فراکشن دارای فعالیت ضد تشنجی (f1) با(Matrix Assisted Laser Desorption/Ionization (MALDI mass تعیین شد.یافته هاعصاره تا دوز 7 گرم بر کیلوگرم هیچگونه سمیتی نداشت. در میان سه مدل تشنجی عصاره و فراکشن f1 در مدل پنتیلن تترازول اثر ضد تشنجی داشتند. در عصاره و فراکشنها میزان قابل توجهی الکالوئید شناسایی شد. در فراکشن f1 یک مولکول با ساختار جدید شناسایی شد.نتیجه گیریکشف یک مولکول ضد تشنجی با ساختمان شیمیایی جدید در دانه نخود نوید بخش یک منبع جدید و ارزان قیمت از داروهای ضد صرع می باشد. با این حال نیاز است تا مطالعات تکمیلی دقیق در زمینه مکانیسم این اثر و رابطه ساختمان-عمل برای این مولکول انجام شود.

صفحات 51-56پیشزمینهنوروپاتی گلوکوماتوز یک نوع مرگ سلولی ناشی از آپوپتوز می باشد. ژن p53 یکی از ژن های نظارتی آپوپتوز است. اخیرا، ارتباط بین ژن کدون 72 ژن p53 با بیماری گلوکوم زاویه باز اولیه (POAG)، در برخی از گروه های قومیتی بررسی شده است. این مطالعه برای اولین بار همراهی بین POAG و پلی مورفیسم کدون 72 ژن P53 را در ایران بررسی کرده است.مواد و روش هاگروهی از 65 بیمار غیر مرتبط مبتلا به POAG (محدوده سنی 12-62 سال، میانگین ± انحراف معیار 17.51 ± 40.16 سال) و 65 نفر به عنوان گروه شاهد (بدون POAG، محدوده سنی 14-63 سال، میانگین ± انحراف معیار 35.64 ± 13.61 سال) انتخاب شدند. در گروه بیماران و شاهد، پلی مورفیسم کدون 72 ژن p53 در اگزون 4 با استفاده از PCR تکثیر شد. قطعات DNA حاصل با استفاده از آنزیم محدودالاثر BstUI هضم شده و از الگوهای هضم برای شناسایی آلل های کدون Pro72Arg مورد استفاده قرار گرفت.یافته هانتایج تفاوت معنی داری در فراوانی آللی و ژنوتیپی کدون Pro72Arg بین بیماران مبتلا به POAG و گروه شاهد نشان داد. احتمال ابتلای بالاتری به بیماری POAG برای آلل(Pro (OR = 2.1, 95% CI=1.2–3.4 و ژنوتیپ (Pro/Pro (OR = 3.9, 95% CI = 0.13-12.7 مشاهده شد.نتیجه گیریفراوانی آلل Pro کدون 72 ژن p53 در بین بیماران POAG نسبت به گروه کنترل (P <0.05) در ایران بیشتر بود. یافته های این مطالعه نشان می دهد که احتمال ابتلا به POAG در بین افراد با ژنوتیپ Pro/Pro بیشتر می باشد. با این حال، مطالعات بیشتری برای تایید این یافته ضروری می باشد.

بررسی همزمان سه ژنوتیپ C1236T, C3435T, G2677T/A در ژن MDR1 جمعیت شهر همدان، غرب ایرانصفحات 57-62پیشزمینهیکی از محدودیت های درمان بیماری های شایعی مانند سرطان، ایجاد مقاومت داروئی است. پلی مورفیسم ها می توانند سطح عملکرد ژن MDR1 که نقش مهمی در مقاومت داروئی دارد را تغییر دهند. در مطالعه حاضر، فراوانی همزمان سه ژنوتیپ C1236T, C3435T, G2677T/A ژن مذکور در نمونه نسبتا بزرگی از جمعیت شهر همدان، با هدف ایجاد یک پایگاه اطلاعاتی، تعیین گردید.روش کار2میلی لیتر خون محیطی از 935 نفر از افراد شرکت کننده گرفته شد و DNA آن استخراج گردید. روش Multiplexed mutagenically separated PCR و در ادامه الکتروفورز بر روی ژل پلی اکریل آمید و رنگ آمیزی با نیترات نقره انجام شد. روش توالی یابی جهت تائید صحت نتایج در مورد 10 نمونه تصادفی انجام گردید. در انتها آلل ها و ژنوتیپ های 933 نمونه (157 مرد و776 زن) تعیین شد.نتایجفراوان ترین آلل ها G2677، 3435T و C1236و فراوان ترین ژنوتیپ ها 2677G/A، 3435C/Tو 1236C/T بودند.درمیان احتمالات ممکن در مورد حضور همزمان ژنوتیپها، شایعترین حالت وجود همزمان سه ژنوتیپ مذکور بود. فراوانی ژنوتیپها در دو گروه مردان و زنان تفاوت معنی داری نداشت.
بحث: نتایج حاصله از این مطالعه شبیه به دیگر مطالعات در ایران بود و مقایسه آن با نتایج سایر گروه های نژادی، شباهت بیشتر با نژادهای آسیائی نسبت به اروپائی ها را نشان داد. از منظر دانش پزشکی شخصی، داده های این تحقیق می تواند در بهبود درمان داروئی بیماری هائی مانند سرطان موثر باشد.

صفحات 63-68مقدمههدف از این پژوهش، مطالعه ی توزیع و تغییرات گلیکوکانژوگیت ها به ویژه قند های انتهایی آن ها در طی تکامل طناب نخاعی موش با استفاده از تکنیک لکتین هیستوشیمی بود.مواد و روش هابرش های بافتی (جنین های 10 تا 16 روزه) فیکس شده با فرمالین برای متد لکتین هیستوشیمی آماده شدند. در این مطالعه، از دو گروه لکتین اختصاصی نشاندار شده با آنزیم horseradish peroxidase استفاده شد: گروه N-acetylgalactosamine شاملDBA, WFA, VVA, SBA و گروه لکتین های متصل به مانوز شامل LTA, UEA-1, OFA. برای رنگ زمینه نمونه ها از آلسین بلو با pH=2.5 استفاده شد.نتایجنتایج ما نشان دادا که تنها لکتین های WFA و OFA واکنش شدیدی با سلول های صفحه کفی نخاع در دوران اولیه و نهایی تکوین نشان دادند. شدیدترین واکنش مربوط به نمونه های بدست آمده از روزهای 14، 15 و 16 جنینی که در معرض لکتین های WFA و OFA بود.نتیجه گیریمطالعه ی حاضر مشخص کرد که تمایز سلولی و مولکولی سازمان دهنده های نخاع یک فرآیند کاملا تنظیم شده است و قندهای انتهایی α-L-fucose, α-D-GalNAc, and α/β-D-GalNAc نقش مهمی را در تکوین جنینی نخاع ایفا می کنند.

Pages 1-16BackgroundKinesin spindle protein (KSP) plays a critical role in mitosis. Inhibition of KSP function leads to cell cycle arrest at mitosis and ultimately to cell death. The aim of this study was to suppress KSP expression by specific small-interfering RNA (siRNA) in Hep3B cells and evaluate its anti-tumor activity.MethodsThree siRNA targeting KSP (KSP-siRNA #1-3) and one mismatched-siRNA (Cont-siRNA) were transfected into cells. Subsequently, KSP mRNA and protein levels, cell proliferation, and apoptosis were examined in both Hep3B cells and THLE-3 cells. In addition, the chemosensitivity of KSP-siRNA-treated Hep3B cells with doxorubicin was also investigated using cell proliferation and clonogenic survival assays.ResultsThe expression of endogenous KSP at both mRNA and protein levels in Hep3B cells was higher than in THLE-3 cells. In Hep3B cells, KSP-siRNA #2 showed a further downregulation of KSP as compared to KSP-siRNA #1 or KSP-siRNA #3. It also exhibited greater suppression of cell proliferation and induction of apoptosis than KSP-siRNA #1 or KSP-siRNA #3; this could be explained by the significant downregulation of cyclin D1, Bcl-2, and survivin. In contrast, KSP-siRNAs had no or lower effects on KSP expression, cell proliferation and apoptosis in THLE-3 cells. We also noticed that KSP-siRNA transfection could increase chemosensitivity to doxorubicin in Hep3B cells, even at low doses compared to control.ConclusionReducing the expression level of KSP, combined with drug treatment, yields promising results for eradicating hepatocellular carcinoma (HCC) cells in vitro. Pages 17-22IntroductionAlthough extensive research has been conducted on lung cancer markers, a singular clinically applicable marker has not been found yet. The objective of this study was to evaluate the sensitivity and the specificity of carcinoembryonic antigen (CEA) mRNA and lung-specific X protein (LUNX) mRNA biomarkers in peripheral blood to detect lung cancer individually and simultaneously.MethodsThirty patients affected by lung cancer and 30 healthy individuals were studied in this research. Three vials of cDNA were made from each sample after taking peripheral blood samples and extracting total RNA. Each sample was examined by the real-time RT-PCR technique. The result from each vial was then compared with the sensitivity of overall marker.ResultsThe CEA mRNA was positive in 24 out of 30 lung cancer patients. Hence, its sensitivity was determined at 80%, differing significantly from that observed in healthy individuals, where 11 positive cases were seen. The overall sensitivity of this marker was significantly associated with positivity in vials 2 and 3 but not in vial 1. The LUNX mRNA was positive in 21 out of 30 patients, indicating 70% sensitivity. This finding significantly differed from that in healthy individuals. The overall sensitivity of this marker was significantly associated with positivity in vials 1 and 3, but not in vial 2. In 93.3% of the patients, at least one positive marker was observed.ConclusionsThe mentioned mRNA could be suggested as sensitive and specific markers in peripheral blood for primary diagnosis of lung cancer.Keywords: Lung neoplasms, RNA, Carcinoembryonic antigen, Sensitivity, specificity

Comparison of Mitochondrial-Related Transcriptional Levels of mitochondrial transcription factor A, Nuclear respiratory factor 1 and cytochrome c oxidase subunit 1 Genes in Single Human Oocytes at Various Stages of the Oocyte MaturationPages 23-28BackgroundThe aim of the current study was to assess the mRNA levels of two mitochondria-related genes, including nuclear-encoded NRF1 (nuclear respiratory factor 1), mitochondrial transcription factor A [TFAM]), and mitochondrial-encoded (cytochrome c oxidase subunit 1 [MT-CO1]) genes in various stages of the human oocyte maturation.MethodsOocytes at the various stages of maturation were isolated from nine infertile women with male factor undergoing in vitro fertilization (IVF)/intra-cytoplasmic sperm injection protocol Mitochondrial-related mRNA levels of target genes were performed by single-cell taqman real time PCR-based assay.Resultsthe expression level of the target genes was low at the germinal vesicle stage (P>0.05). Although the mRNA level of NRF1gene remained stable in metaphase I, the mRNA level of TFAM and MT-CO1 increased significantly (P<0.05).In metaphase II, the expression level of all genes increased compared to metaphase I (P<0.05).ConclusionThe overexpression levels of NRF1, TFAM, and MT-CO1genes are related to the oocyte maturation. Therefore, the current study could be used clinically to improve the success rate of IVF.

Pages 29-34BackgroundBrain hypoxia-ischemia is a human neonatal injury that is considered a candidate for stem cell therapy.MethodsThe possible therapeutic potential of human umbilical cord blood (HUCB) stem cells was evaluated in 14-day-old rats subjected to the right common carotid occlusion, a model of neonatal brain hypoxia-ischemia. Seven days after hypoxia-ischemia, rats received either saline solution or 4 × 105 HUCB cells i.v. Rats in control group did not receive any injection. After two weeks, rats were assessed using two motor tests. Subsequently, rats were scarified for histological and immunohistochemical analyses.ResultsOur immunohistochemical findings demonstrated selective migration of the injected HUCB cells to the ischemic area as well as reduction in infarct volume. Seven days after surgery, we found significant recovery in the behavioral performance in the test group (12.7 +/- 0.3) compared to the sham group (10.0 +/-0.05), a trend which continued to day 14 (15.3 ± 0.3 vs. 11.9 ± 0.5, P<0.05). Postural and motor asymmetries at days 7 and 14 in the test group showed a significant decrease in the percentage of right turns in comparison to the sham group (75% and 59% vs. 97% and 96%, P<0.05).ConclusionThe results show the potential of HUCB stem cells in reduction of neurologic deficits associated with neonatal hypoxia-ischemia.Keywords: Hypoxia, Ischemia, Nerve cell, Umbilical Cord Blood

Pages 35-44BackgroundCD4+ and CD8+ T cells are the main types of lymphocytes in cell-mediated immunity and play a central role in the induction of efficient immune responses against tumors. The frequencies of T cell subtypes in the peripheral blood and tumor tissues, and draining lymph nodes (dLN) can be considered as useful markers for evaluation of the immune system in cancers.MethodsIn this study, the frequencies of CD4+ and CD8+ T cells in blood, tumor tissues, and dLN samples of breast cancer patients were compared with each other and with similar tissues from normal individuals. Immunophenotyping was carried out by flow cytometry and the expression levels of CXCL10, granzyme B, and mammaglobin were evaluated by real-time PCR.ResultsIn the peripheral blood, there were no differences in the T cell subsets between the patients and the normal individuals. The frequency of CD8+ T cells was significantly higher in tumor tissue than normal breast tissues while granzyme B expression was similar. Based on mammaglobin expression levels, dLN have been classified into micro- and macro-metastatic dLN. We found significantly lower frequency of CD4+ in macro-metastatic dLN than micro-metastatic dLN. CD8+ frequency was similar in both dLN; however, granzyme B expression was higher in micro-metastatic ones. There was not any significant difference in CXCL10 expression between the two types of dLN.ConclusionsBased on our results, although the tumor does not affect the systemic immunity, tumoral cells affect the local immune system in the tumoral tissues and the metastatic dLN.Keywords: Breast neoplasms_CD4+ T lymphocyte_CD8+ T lymphocytes_Granzymes_CXCL10

Pages 45-50BackgroundCicer arietinum (Chickpea) is one of the most important harvests in the world with high nutritional value. Lack of essential oils in the seeds of Chickpea is an advantage in search for drug-like molecules with less toxicity. We evaluated anticonvulsant effect of C. arietinum in common animal models of epilepsy.MethodsDichloromethane extract was obtained from C. arietinum seeds by percolation. Acute toxicity of the extract was assessed in mice. Protective effect of the extract was examined against tonic seizures induced by maximal electroshock (MES; 50 mA, 50 Hz, 1 s) in mice, clonic seizures induced by pentylenetetrazole (PTZ; 60 mg/kg, i.p.) in mice, and electrical kindling model of complex partial seizures in rats. The extract was fractionated by n-hexane to f1 and f2 fractions. The extract and fractions underwent phytochemical analysis by thin layer chromatography. The active anticonvulsant fraction, f1, was subjected to matrix assisted laser desorption/ionization (MALDI) mass analysis.ResultsThe crude extract had neither toxicity up to 7 g/kg nor protective activity in MES and kindling models. However, it significantly inhibited clonic seizures induced by PTZ. f1 fraction mimicked protective effect of the extract. Phytochemical screening revealed the presence of considerable amount of alkaloids in the extract and fractions. Moreover, a novel structural class was detected in f1 fraction.ConclusionFinding an anticonvulsant molecule pertaining to a new structural class in the seeds of C. arietinum promises an effective and inexpensive source of antiepileptic medication. Pages 51-56BackgroundGlaucomatous neuropathy is a type of cell death due to apoptosis. The p53 gene is one of the regulatory genes of apoptosis. Recently, the association between the p53 gene encoding for proline at codon 72 and primary open-angle glaucoma (POAG) has been studied in some ethnic groups. This study is the first association analysis of POAG and p53 codon 72 polymorphism in Iranian patients.MethodsA cohort of 65 unrelated patients with POAG (age range from 12-62 years, mean ± SD of 40.16 ± 17.51 years) and 65 unrelated control subjects (without glaucoma, age range of 14-63 years, mean ± SD of 35.64 ± 13.61 years) were selected. In Iranian POAG patients and normal healthy controls, the p53 codon 72 polymorphism in exon 4 was amplified using polymerase chain reaction. The amplified DNA fragments were digested with the BstUI restriction enzyme, and the digestion patterns were used to identify the alleles for the polymorphic site.ResultsComparisons revealed significant differences in allele and genotype frequencies of Pro72Arg between POAG patients and control group. A higher risk of POAG was associated with allele Pro (OR = 2.1, 95% CI = 1.2–3.4) and genotype Pro/Pro (OR = 3.9, 95% CI = 0.13-12.7).ConclusionThe p53 Pro72 allele was more frequent in Iranian POAG patients than in the control group (P<0.05). The present findings show that the individuals with the Pro/Pro genotype may be more likely to develop POAG. Simultaneous Analysis of Multidrug Re sistance 1(MDR1) C3435T, G2677T/A, and C1236T genotypes in Hamadan City Population, West of IranPages 57-62BackgroundOne of the limitations in the treatment of common diseases such as cancer chemotherapy is development of multidrug resistance (MDR). Polymorphisms could alter the expression level of MDR1 gene, which plays an important role in MDR. In this research, the frequency of C3435T, C1236T, and G2677T/A polymorphisms of MDR1 gene was investigated in a large group of population from Hamadan city to provide a sample data resource.MethodsPeripheral blood (2 ml) was taken, and DNA extraction was carried out. Multiplexed mutagenically separated PCR, which was followed by polyacrylamide gel electrophoresis and silver staining, was applied to detect the mentioned polymorphisms in 935 individuals. Sequencing performed for confirmation of gel electrophoresis resulted in 10 random cases. In total, alleles and genotypes of 933 persons (776 women and 157 men) were determined.ResultsThe most frequent alleles of the polymorphisms were: 3435T, C1236, and G2677. The most frequent genotypes were: 3435C/T, 1236C/T, and 2677G/A, and their concurrent presence was also found as the most frequent simultaneous genotypes. There was not any meaningful difference among the prevalence of these genotypes in groups of men and women.ConclusionOur results were close to those of other studies performed in Iran and compared to the other ethnic groups, which showed more similarity to Asian peoples than Europeans. Pages 63-68BackgroundThe aim of this research was to study the distribution and changes of glycoconjugates particularly their terminal sugars by using lectin histochemistry during mouse spinal cord development.MethodsFormalin-fixed sections of mouse embryo (10-16 fetal days) were processed for lectin histochemical method. In this study, two groups of horseradish peroxidase-labeled specific lectins were used: N-acetylgalactosamine, including Dolichos biflorus, Wisteria floribunda agglutinin (WFA), Vicia villosa, Glycine max as well as focuse-binding lectins, including tetragonolobus, Ulex europaeus, and Orange peel fungus (OFA). All sections were counterstained with alcian blue (pH 2.5).ResultsOur results showed that only WFA and OFA reacted strongly with the floor plate cells from early to late embryonic period of developing spinal cord. The strongest reactions were related to the 14, 15, and 16 days of tissue sections incubated with OFA and WFA lectins.ConclusionThe present study demonstrated that cellular and molecular differentiation of the spinal cord organizers is a wholly regulated process, and α-L-fucose, α-D-GalNAc, and α/β-D-GalNAc terminal sugars play a significant role during the prenatal spinal cord development.Keywords: Development, Glycoconjugates, Spinal cord
