Isolation of crude oil degrading Bacillus from Isfahan Oil Refinery contaminated soil

In the oil-producing countries, this substance considered as one of the most significant environment contamination. Release of oil in the water and soil can cause damage to living organisms. In this regard, the current study investigated isolation stages of crude oil parser bacillus from contaminated soil of Isfahan oil refinery. Soil samples were collected from Isfahan refinery, then they cultured at MSM culture medium containing 1% crude oil as the sole carbon source. By cultivation of bacteria on blood agar, the bacteria with beta hemolysis were isolated, surface tension test was performed to detect Biosurfactants produced by bacillus and emulsification test was conducted for selected bacillus. Bacillus were identified by biochemical tests and molecular techniques. Finally, using gas chromatography mass spectrometer (GC-MS) and by spectrophotometric method, oil removal rate by bacillus was obtained in compare to control sample. Among the 12 strains isolated bacillus, 4strains had beta hemolytic. 2 strains surface tension was acceptable, mN/m 36 and 34 respectively. 2strains emulsification ability was measured 65% and 75% respectively. And by two strains molecular analysis were identified as Bacillus cereus and Bacillus thuringiensis.
