Tetraploid Induction by Colchicine in Yagoti Grape (Vitis vinifera var.Yagoti)

Abstract:
Growth performance and crop increasing based on production of seedless grape and berry growth using autotetraploid is seeing necessary. The current study was conducted to investigate on increasing grape berries size, marketing and yield, and to determine the best concentration and time length of colchicines. The experiment was carried out in spring season in a Takestan grape researches station. In The first decade of March, favorite grape rootstocks (Vitis vinifera var. Yagouti) were selected and labeled. It was in a factorial RCBD with 4 replications. Colchicine's concentrations were 0.1 %, 0.3 %, 0.5 %, 0.7 %, 0.9 % and 1.1 % with 4 levels of time length 24, 48, 72 and 96h. Colchicine was applied using cotton in different times on closed buds. Leaves sample were collected for cytological study and tetraploide conformation 60 days after treatments. ANOVA table and means comparisons were administrated by DMRT test using SAS software. Colchicines concentration with 0.9% and 1.1% via 72 and 96h were the beast treatment for autotetraploidy induction in Vitis vinifera var. Yagoti respectively. However, samples were taken for cutting from the trees which were treated by colchicines in autumn. Samples were collected to cytological study after rooting. Tetraploid characters were observed in 0.9% and 1.1% concentration via 72 and 96h treatments respectively but the other treatments identified as a diploid. In these two treatments significant differences were observed in the anatomical structure of the cellular structure of petioles, stems, leaves and roots were observed in different treatments compared to non-treated and other treated samples.
