Distribiution of Potato Virus Y in Jahrom area

In this study three hundred fifty samples collected from diseased potato, tomato and eggplant plants that showed virus suspected symptoms such as: mosaic and yellowing of leaves, necrotic spots on veins and stunting from Jahrom region of Fars province. The samples were detected by using enzyme linked immunosorbent assay (DAS-ELISA) method with polyclonal antibodies for serological distinguishing of Potato Virus Y (PVY). Almost sixty samples collected from potato were infected by PVY in this phase. For molecular diagnostic of the virus, RT-PCR assay were used for infected samples with primer pairs of P1 and P2. These primers could amplify the coat protein region of virus genome. Eventually PCR products of two samples were sequenced and compared with 19 sequences available in GenBank using Blast search. The MEGA6 software and UPGMA method were used to draw phylogenetic tree. The drawn tree showed that the isolates of jahrom-Iran and the most isolates of Europe were placed in separate clusters.This results showed that in this area, virus infected only found in potato plants so it is possible that crop infections were related to use of infected seed tubers. This is the first report of in Jahrom area.
