افزایش شیوع بیماری های متابولیک (مثل: دیابت و چاقی) به طور جدی سلامت انسان و ایمنی زندگی را در سراسر جهان تحت تاثیر قرار داده است. یافتن داروهای موثر با ترکیبات طبیعی و فعالیت ورزشی برای درمان بیماری های متابولیک از اهمیت بالایی برخوردار است. هدف از پژوهش حاضر بررسی اثر مصرف بربرین کلراید با تمرین هوازی بر بیان ژن Nrf2، HO-1 و PPARγ کبدی موش های صحرایی دیابتی با استرپتوزوتوسین (STZ) می باشد.
روش کاردر این مطالعه تجربی، 32 موش صحرایی نر ویستار به طور تصادفی به چهار گروه (8=n): دیابت (DM)، دیابت-بربرین (BDM)، دیابت- تمرین هوازی (TDM)، دیابت- تمرین هوازی- بربرین (TBDM) تقسیم شدند. دیابت با تزریق STZ در موش های نر القا شد. بربرین کلراید (mg/kg/day 30) یک بار در روز به صورت خوراکی با گاواژ خورانده شد. گروه های تمرین به مدت شش هفته برنامه تمرین هوازی فزاینده (18-10 متر در دقیقه، 40-10 دقیقه در روز، پنج روز در هفته) را روی تردمیل انجام دادند.
یافته هابیان Nrf2 و HO-1 در گروه های BDM (به ترتیب =0.027p و p= 0.038)، TDM (به ترتیب p=0.022 و p=0.028) و TBDM (به ترتیب p=0.000 و p=0.000) افزایش معنی داری داشت. این افزایش در گروه های TBDM نسبت به BDM (به ترتیب p=0.034 و p=0.034) و TDM (به ترتیب p=0.043 و p=0.046) نیز مشاهده شد. PPARγ در گروه های TDM (p=0.046) و TBDM (p=0.001) افزایش معنی داری داشت.
نتیجه گیریبربرین کلراید همراه با فعالیت ورزشی احتمالا از طریق تنظیم افزایشی مسیر Nrf2/HO-1 و PPARγ می تواند آسیب کبدی ناشی از STZ را مهار کند.

فعالیت بدنی یکی از عوامل تحریک ترشح فاکتورهای رشد فاکتورهای رشد عصبی در بیماران مبتلا به اماس میباشد. بنابراین هدف از تحقیق حاضر بررسی تاثیر 12 هفته تمرین پیلاتس و مصرف زنجبیل بر مقادیر سرمی BDNF و TNF-α در زنان مبتلا به بیماری اماس بود.
روش کارچهل زن مبتلا به اماس از بین 85 زن مراجعهکننده به اداره بهزیستی شهرستان ارومیه با دامنه سنی 35-30 سال در قالب یک طرح نیمه تجربی با پیش آزمون و پس آزمون، به صورت تصادفی انتخاب شدند و در چهار گروه 10 نفره تمرین+ مکمل، تمرین+ دارونما، مکمل زنجبیل و گروه کنترل تقسیم شدند. گروه های مداخله تمرینات پیلاتس را به مدت 12 هفته، هفته ای 3 جلسه 60 دقیقه ای انجام دادند. و گروه های مکمل، روزانه سه کپسول یک گرمی زنجبیل مصرف کردند. به منظور تحلیل داده ها، از تحلیل کوواریانس با استفاده از نرم افزار SPSS-22به کار رفت.
یافته هانتایج تحقیق حاضر نشان داد که تمرینات ورزشی به همراه مصرف زنجبیل باعث افزایش BDNF در گروه های مداخله در مقایسه با گروه کنترل بود ولی این افزایش در گروه تمرین+مکمل نسبت به دیگر گروه های مداخله بیشتر بود (0/05>p)، همچنین نتایج نشان داد که میزان TNF-α در گروه های مداخله نسبت به گروه کنترل کاهش معنی داری داشت (0/05>p).
نتیجه گیرییافته ها نشان داد که مصرف زنجبیل به عنوان یک روش غیرتهاجمی می تواند اثر مثبتی بر افزایش مقادیر BDNF و کاهش مقادیر TNF-αدر زنان مبتلا به اماس داشته باشد.

اثر تمرینات استقامتی و ازن درمانی بر بیان ژن FLS و VEGF در بافت زانو در موشهای مبتلا به استئوآرتریتصفحات 318-327زمینه و هدف
استیوآرتریت یک بیماری است که در نتیجه تخریب غضروف مفصلی بروز میکند و منجر به درد و سفتی مفاصل میگردد. هدف از تحقیق حاضر بررسی اثر تمرینات استقامتی و ازن درمانی بر بیان ژن FLS و VEGF در بافت زانو موش های مبتلا به استیوآرتریت زانو میباشد.
روش کارآزمودنی ها شامل موش های صحرایی نر ویستار بودند که پس از سازگاری دو هفته ای به صورت تصادفی به 5 گروه شامل گروه های: کنترل- سالم، کنترل- بیمار، بیمار- ازن، بیمار- ورزش و بیمار- ورزش- ازن تقسیم شدند. برای سازگاری موش ها با ورزش و تردمیل، قبل از ورزش، برنامه پیاده روی انجام گردید. برنامه تمرین شامل 30 دقیقه دویدن روی تردمیل بدون شیب و با سرعت 16 متر در دقیقه برای هفته اول بود و هر هفته یک متر بر دقیقه اضافه شد تا در هفته هشتم به 22 متر بر دقیقه رسید. ازن از طریق خط مفصل تیبیوفمورال با غلظت 20 میکروگرم بر میلی لیتر، یک بار در هفته و در ساعت 9 صبح و به مدت 3 هفته به زانو تزریق شد. سطح بیان ژنهای FLS و VEGF توسط Real Time PCR اندازه گیری شد. برای تعیین تفاوت بین گروهها از روش ANOVA و توکی استفاده شد.
یافته هانتایج حاصل از تحلیل آماری نشان داد که سطح بیان ژن FLS و VEGF در موش های مبتلا به استیوآرتریت افزایش معناداری داشت. همچنین نشان داده شد که تمرینات بدنی و ازن درمانی موجب روند کاهشی سطح تغییرات بیان ژن FLS و VEGF در موش های مبتلا به استیوآرتریت شد.VEGF و گیرنده های آن در غضروف OA بیان شده است و این احتمال را نشان می دهد که VEGF برای از بین بردن غضروف مفصلی OA از طریق افزایش تولید MMP نقش دارد.
نتیجه گیرینتایج این مطالعه امیدوارانه در مورد نقش VEGF به عنوان یک نشانگر تشخیصی است. VEGF می تواند در تغییرات اولیه OA شرکت کند و یک روش درمانی با تعدیل تولید VEGF می تواند یک امکان برای آینده باشد.

Foxo1 یک فاکتور رونویسی در سیگنالینگ انسولین در کبد می باشد. عملکرد Foxo1 بر توانایی انسولین در تنظیم تولید گلوکز کبدی و هموستاز گلوکز، مهم می باشد. مطالعه حاضر با هدف تعیین اثر 6 هفته تمرین مقاومتی بر بیان Foxo1 در سلول های کبدی و همچنین سطوح گلوکز و انسولین در موش های صحرایی دیابتی نوع 2 انجام گرفت.
روش کارجامعه آماری این مطالعه تجربی را کلیه موش های صحرایی نر ویستار تشکیل دادند که از بین آنها 14 سر موش صحرایی 10 هفته ای با وزن 20±220 گرم خریداری شدند. در ادامه موش های صحرایی مورد مطالعه تحت اثر 6 هفته رژیم غذایی پرچرب و تزریق STZ دیابتی نوع 2 شدند و به شیوه تصادفی در دو گروه (کنترل و مقاومتی) قرار گرفتند. سپس گروه تمرین در یک دوره تمرین مقاومتی به مدت 6 هفته به تعداد 5 جلسه در هفته در قالب 4 ست و 5 تکرار در هر ست در صعود از نردبان 26 پله ای به ارتفاع 1 متر با شیب عمودی 80 درصد، شرکت کردند. نهایتا 48 ساعت پس از آخرین جلسه تمرین، سطوح ناشتایی گلوکز و انسولین و بیان Foxo1 در سلول های کبدی هر دو گروه اندازه گیری و توسط آزمون تی مستقل با یکدیگر مقایسه شدند (5% = a).
یافته هاتمرینات مقاومتی به بهبود سطوح گلوکز ناشتا در مقایسه با گروه کنترل منجر شد (0/001>p). سطوح انسولین سرم به میزان معنی داری افزایش یافت (0/042 =p) و بیان Foxo1 در سلول های کبدی در گروه مقاومتی در مقایسه با گروه کنترل به میزان معنی داری کاهش یافت (0/023 =p).
نتیجه گیریبر پایه این یافته ها، احتمالا کاهش گلوکز در گروه تمرین را می توان به کاهش بیان Foxo1 در سلول های کبدی در واکنش به تمرینات مقاومتی نسبت داد.

اختلالات میتوکندریایی از عوامل اصلی خطرزا در ایجاد بیماری های عصبی مرتبط با پیری بشمار می روند. از سوی دیگر، ورزش هوازی اثرات مفیدی بر سلامت مغز و عملکرد شناختی دارد و پویایی میتوکندریایی را بهبود می بخشد. از این رو، هدف از مطالعه حاضر، بررسی اثر 4 هفته تمرین هوازی بر عملکرد یادگیری و حافظه فضایی و پویایی میتوکندریایی در بافت هیپوکمپ رت های پیر بود.
روش کاربدین منظور، تعداد 14 سر رت نر بالغ نژاد ویستار 20 ماهه به روش تصادفی ساده به 2 گروه ورزش هوازی (تعداد = 7 سر) و گروه کنترل (تعداد = 7 سر) تقسیم شدند. گروه ورزش 4 هفته تمرین دویدن روی نوارگردان (5 روز در هفته با سرعت 10 الی 15 متر بر دقیقه) را اجرا نمودند. چهل و هشت ساعت پس از آخرین جلسه تمرینی حیوانات تحت آزمون رفتاری قرار گرفتند. بیست و چهار ساعت بعد از آزمون رفتاری تمامی رت ها کشته شدند و بافت هیپوکمپ استخراج شد. بیان mRNA ژن های OPA1، Mfn2 وDrp1 با استفاده از روش RealTime-PCR مورد سنجش قرار گرفت. برای تجزیه و تحلیل آماری از آزمون t مستقل استفاده شد.
نتایجتمرین هوازی در حیوانات پیر عملکرد یادگیری و حافظه فضایی را بهبود بخشید و میزان بیان ژن OPA1 هیپوکمپ را افزایش و میزان بیان ژنDrp1 را در مقایسه با گروه کنترل کاهش داد (0/01 ≥ p).
نتیجه گیریبه نظر می رسد که تمرین هوازی می تواند عملکرد میتوکندری های مغز را از طریق تعدیل فرایندهای همجوشی و شکافت بهبود بخشیده و به عنوان یک روش غیردارویی موثر برای مقابله با اختلالات یادگیری و حافظه در پیری در نظر گرفته شود.

کبد یکی از بافت های مهم بدن و محل متابولیسم اکثر داروها و سموم می باشد. بنابراین هرگونه آسیب به کبد باعث اختلال در عملکرد آن و بوجود آمدن بیماری های مختلفی می شود. از طرفی با توجه به گرایش روز افزون به مصرف گیاهان دارویی، در این مطالعه اثرات محافظت کبدی ترکیب عصاره آبی گیاه رازک (Humulus lupulus) بر آسیب کبدی القا شده توسط تتراکلرید کربن در موش صحرایی مورد بررسی قرار گرفت.
روش کار36 سر موش صحرایی نژاد ویستار انتخاب و به 6 گروه 6 تایی به شکل تصادفی تقسیم شدند. گروه های 1 و 2 به مدت 14 روز سرم فیزیولوژی و گروه 3، 4 و 5 به ترتیب دوز200،100،50 میلی گرم بر کیلوگرم عصاره رازک را به مدت 14 روز دریافت کردند. گروه6: به مدت 14 روز دوز 100 میلی گرم بر کیلوگرم سیلیمارین (داروی استاندارد) به شکل خوراکی دریافت نمود. همه گروهها بجز گروه1 در روز 14، 1 میلی لیتر بر کیلوگرم تتراکلریدکربن دریافت کردند. پس از بیهوشی حیوانات، نمونه خون آنها به منظور آزمایشات بیوشیمیایی مورد استفاده قرار گرفت.
یافته هاپیش درمان با عصاره گیاه رازک به طور معنی دار فعالیت سرمی آنزیم های کبدی و نیز سطح تریگلیسیرید را کاهش و لیپوپروتیین با چگالی بالا (HDL) را افزایش داد (0/05>p) اما تاثیر قابل توجهی بر روی آنزیم گاماگلوتامیل ترانسفراز، کلسترول تام و لیپوپروتیین با چگالی پایین (LDL) نداشت.
نتیجه گیرینتایج این مطالعه نشان داد که عصاره گیاه رازک میتواند کبد را از آسیب های ناشی از عوامل دارویی و سمی حفظ کرده و اثرات مفید بر پروفایل لیپیدی داشته باشد.

تاثیر تمرین استقامتی با شدت متوسط بر بیان فاکتورهای آنژیوژنز و استرس اکسیداتیو قلب در موش های صحراییصفحات 361-371زمینه و هدف
با توجه به نقش احتمالا مثبت تمرین ورزشی بر وضعیت اکسیدانی تام (TOS) و سیستم آنتی اکسیدانی تام (TAS) و اثر متقابل آنها بر روند پروسه آنژیوژنز، هدف از مطالعه حاضر بررسی تاثیر یک دوره تمرین استقامتی با شدت متوسط بر میزان بیان ژن فاکتور رشد اندوتلیال عروق- (VEGF-B) Bوآنژیوپوییتین-1 (ANGPT-1) و مقادیر TOS و TAS بافت قلب در موش های صحرایی نر بود.
روش کاردر این پژوهش 20 سر موش نر صحرایی نژاد ویستار به دو گروه مساوی بدین شرح تقسیم شدند: 1- گروه تمرین و 2- گروه کنترل که هیچ گونه فعالیت ورزشی بر روی آن ها انجام نشد. موش های گروه تمرین از هفته دوازدهم زندگی به مدت شش هفته و هر هفته 5 جلسه تمرین استقامتی انجام دادند. 24 ساعت پس از آخرین جلسه تمرین برای بررسی اثر تمرین، نمونه های بافت قلب استخراج گردید. برای مقایسه بین گروهی و اختلاف معنادار بین گروه ها از آزمون آماری t مستقل با سطح آماری 0/05˂p استفاده شد.
یافته هانتایج نشان داد که در گروه تمرین نسبت به گروه کنترل میزان بیان ژن VEGF-B و ANGPT-1 و مقادیر TAS افزایش معنادار 0/001 =pو مقادیر TOS کاهش معنادار =0/008pداشتند.
نتیجه گیریباتوجه به نتایج حاصل از پژوهش حاضر به نظر می رسد که تمرین استقامتی با شدت متوسط با افزایش فاکتورهای مرتبط با آنژیوژنز بافت قلب موش ها و بهبود TAS و کاهش TOSدر پیشگیری از بیماری های قلبی- عروقی می تواند موثر باشد.

کاربری جدید دارویی عبارت است از کاربرد برخی از داروها برای درمان یا کنترل علایم بیماریهای دیگر. مزیت اصلی این تکنیک در انتخاب یک دارو به عنوان ترکیب الگو است که به دلیل عدم وجود مشکلات جدی درفارماکوکینتیک، بهینه سازی فارماکودینامیک را هدف قرار میدهد. با عنایت به اهمیت موضوع، پژوهش حاضر به آنالیز مجازی کاربری جدید برخی از دسته های دارویی با هدف دستیابی به فارماکوفورهای بالقوه ضد لیشمانیا اختصاص یافت.
روش کارساختار سه بعدی تعدادی از گیرنده های پروتیینی در انگل لیشمانیا بر اساس گزارشات مقالات معتبر از بانک پروتیینی بروک هاون (PDB) بدست آمدند تا کمپلکس آنها با ساختارهای منتخب دارویی با استفاده از تکنیک داکینگ مولکولی بررسی گردند. آنالیز کیفی و کمی الگوهای برهمکنش در کمپلکس های دارو- گیرنده داک شده، داروهایی که قدرت اتصال بالاتری به گیرنده داشتند را ارایه نمود و در نهایت الگوهای ساختاری یا فارماکوفورهای فرضی ضد لیشمانیا بر اساس ساختارهای دارویی برتر ارایه شدند.
یافتهها: بیشترین انرژی آزاد اتصال برای ناتگلینید در اتصال به آنزیم فارنزیل دی فسفات سینتاز (kcal/mol 13/30- ΔGb) میباشد. در ترکیبات استروییدی، نورژسترل (kcal/mol 9/48-ΔGb ) و تستوسترون (kcal/mol 08/5-ΔGb ) قابلیت اتصال آنزیمی بالاتری نشان دادند و Arg82 نقش مهمی در برقراری پیوندهای هیدروژنی داشت. در الگوی ساختاری حلقهای جوش خورده، میرتازاپین بیشترین انرژی آزاد در اتصال به آنزیم داکسی اوریدین تری فسفات از خود نشان داد (kcal/mol 8/64-ΔGb). در کاربامازپین، استخلاف آمیدی در حلقه میانی زمینه را برای برقراری دو برهمکنش هیدروژنی موثر با اسیدهای آمینه Gln21 و Asn25 داکسی اوریدین تری فسفات فراهم نموده است.نتیجه گیریبر اساس آنالیز الگوهای اتصال، در ساختارهای دارویی استروییدی و سه حلقه ای جوش خورده، امکان طراحی مولکولهایی که در آنها پتانسیل مهار همزمان چندین گیرنده پاتوژنی وجود داشته باشد، وجود دارد.

ABCA1 نقش مهمی در بیوسنتز HDL و هومیوستاز کلسترول سلولی ایفا میکند و بهواسطه این نقش در جلوگیری از تشکیل پلاک های آترواسکلروتیک موثر است. هدف از انجام پژوهش حاضر بررسی تاثیر تمرینات ترکیبی و بازتوانی بر بیان ژن ABCA1 لنفوسیتهای خون و نیمرخ لیپیدی مردان میانسال پس از عمل بای پس عروق کرونر بود.
روش کاردر این مطالعه نوع نیمه تجربی 45 نفر از مردان میانسال (میانگین سنی: 4/7 ±55/58 سال) تحت عمل جراحی بای پس به روش نمونهگیری در دسترس انتخاب و به شکل تصادفی در سه گروه تمرین ترکیبی مقاومتی و هوازی (15نفر)، بازتوانی (15نفر) و کنترل (15نفر) تقسیم شدند. تمرینات ترکیبی شامل تمرینات مقاومتی و هوازی و تمرینات بازتوانی شامل تمرینات هوازی به مدت 8 هفته (هر هفته 3جلسه) بود. نمونه خون در هفتههای صفر و هشت از تمامی افراد جهت سنجش متغیرهای بیان ژن ABCA1، HDL و LDL گرفته شد. جهت تجزیه و تحلیل آماری از آزمونهای کروسکال والیس، آنوای یکطرفه و تعقیبی توکی با استفاده از نرمافزار SPSS و سطح معنیداری 0/05 ≤ p استفاده شد.
یافته هانتایج نشان داد هشت هفته تمرینات ترکیبی و تمرینات بازتوانی باعث افزایش بیان ژن ABCA1 (بهترتیب؛ 0/001= p و 0/001 = p و سطح HDL (به ترتیب؛ 0/001= p و 012/ 0= p پلاسما و کاهش سطح LDL (به ترتیب؛ 0/001= p و 0/001 = p پلاسما شده است. همچنین اختلاف معناداری بین سه گروه در مقادیر ABCA1و HDL به نفع گروه تمرین ترکیبی وجود داشت.
نتیجهگیریبه نظر میرسد انجام تمرینات ترکیبی میتواندبهطور موثرتری نسبت به تمرینات بازتوانی باعث افزایش بیشتر بیان ژن ABCA1 و سطح HDL پلاسما و کاهش سطح LDL در مردان میانسال پس از عمل بای پس عروق کرونر شود.

سرطان پستان شایعترین بیماری و مهمترین عامل مرگ و میر زنان در سراسر دنیا به شمار میرود. هدف مطالعه حاضر تعیین تاثیر پنج هفته تمرین استقامتی به همراه مکمل یاری کورکومین بر بیان ژنهای اینتراتوموری AngiomiR-126 و Angiopoietin-1 در موشهای ماده مبتلا به سرطان پستان بود.
روشکارپژوهش حاضر از نوع تجربی بود.40 سر موش ماده آزمایشگاهی نژاد بالبسی پس از پیوند سرطان پستان 4T1، به چهار گروه کنترل، تمرین استقامتی، کورکومین، تمرین استقامتی- کورکومین تقسیم شدند. گروه تمرین استقامتی و تمرین- کورکومین به مدت 5 هفته (پنج روز در هفته) روزانه 40 دقیقه با شدت 65-60 درصد vVo2peak و شیب صفر درجه بر روی نوار گردان دویدند. گروه کورکومین و تمرین- کورکومین نیز به مدت پنج هفته و شش روز در هفته تحت گاواژ کورکومین قرار گرفتند. 24 ساعت بعد از آخرین جلسه تمرینی، جداسازی بافت سرطان انجام شد. بیان ژنهای miR-126 و Angiopoietin-1 به روش qReal time-PCR در بافت سرطان پستان سنجیده شد. دادهها نیز با آزمون تحلیل واریانس یک طرفه مورد بررسی قرار گرفتند.
یافتههانتایج پژوهش حاضر مهار رشد سرطان پستان، افزایش معنا دار بیان ژن miR-126 (0/001>p) و کاهش معنادار بیان ژن Angiopoietin-1 (0/001>p) در بافت سرطان گروه تمرین نسبت به گروه کنترل را نشان داد. همه این تغییرات در گروه تمرین- کورکومین نسبت به گروههای مداخله دیگر به طور معناداری بیشتر بود (0/001>p).
نتیجه گیریبه نظر میرسد پنج هفته تمرین استقامتی به همراه مکمل یاری کورکومین احتمالا با تاثیر برآیندی باعث کاهش بیشتر رشد توده سرطان پستان از طریق مهار موثرتر محور miR-126/Angiopoietin-1 نسبت به مداخلههای دیگر به تنهایی میشود.

در طی دو دهه گذشته، تحقیقات زیادی برای درک تاثیر ژنتیک بر عملکرد ورزشی تمرکز داشتهاند. توالییابی کل اگزوم پتانسیل زیادی برای شناسایی واریانت های ژنتیکی مرتبط با عملکرد جسمانی دارد، اما با این وجود در تحقیقات ژنتیک ورزشی هنوز از این تکنیک جدید استفاده نشده است. بنابراین هدف از نگارش مقاله حاضر گزارش واریانتهای جدید در ژن های درگیر در قدرت و هایپرتروفی عضلانی (ACE، NOS3، IGF1R و IL-6) در یکی از قهرمانان وزنه برداری جهان با استفاده از روش توالی یابی کل اگزوم بود.
روش کارپس از اخذ رضایت نامه کتبی از یکی از ورزشکاران عضو تیم ملی وزنه برداری مردان، 5 سی سی خون دریافت شد و پس از استخراج DNA از نمونه خون، با استفاده از روش توالی یابی کل اگزوم مورد بررسی قرار گرفت.
یافته هانتایج حاصل از توالی یابی کل اگزوم نشان دهنده وجود واریانت بد معنی (99454613G>A) در ژن IGF1R، پلیمورفیسم (22771156C>T) در ژن IL-6، پلی مورفیسم های (61564052A>G و 61573761T>C) در ژن ACE و پلیمورفیسم های (150695726T>C و 150704250C>G) در ژن NOS3 بود.
نتیجه گیرینتایج تجزیه و تحلیل توالی یابی کل اگزوم واریانت و پلی مورفیسم هایی را در ژن های مرتبط با هایپرتروفی و قدرت عضلانی در این وزنه بردار نخبه نشان داد. تحقیقات دیگری با حجم نمونه بیشتر جهت بررسی ارتباط این واریانت و پلی مورفیسم ها با عملکرد ورزشکاران قدرتی مورد نیاز است.

The increased incidence of metabolic diseases (e.g., diabetes and obesity) has seriously affected human health and life safety worldwide. It is of great significance to find effective drugs from natural compounds and exercise to treat metabolic diseases. The aim of this study was to evaluate the effect of berberine chloride with aerobic training on Liver Gene Expression Nrf2, HO-1 and PPARγ in Streptozotocin (STZ)-induced diabetic rats.
MethodsIn this experimental study, 32 male Wistar rats were randomly divided into four groups (n=8): Diabetes (DM), Diabetes-Berberine (BDM), Diabetes-Aerobic Training (TDM), Diabetes-Aerobic Training -Berberine (TBDM). Diabetes was induced by injection of STZ in male rats. Berberine chloride (30 mg/kg/day) were administered orally, by gavage, once a day. Training groups have performed a progressive aerobic running program (at 10-18 m/min, 10-40 min/day, and 5 days/week) on a motor-driven treadmill for six weeks.
ResultsThere was a significant increase in Nrf2 and HO-1 expression in BDM in BDM (p=0.027 and p=0.038, respectively), TDM (p=0.022 and p=0.028, respectively) and TBDM (p=0.000 and p=0.000, respectively). This increase was also observed in TBDM groups compared to BDM (p=0.034 and p=0.034, respectively) and TDM (p=0.043 and p=0.046, respectively). PPARγ was significantly increased in TDM (p=0.046) and TBDM (p=0.001) groups.
ConclusionsBerberine chloride combination with exercise may possibly inhibit STZ-induced liver damage through up-regulating the Nrf2/HO-1 pathway and PPARγ.

Physical activity is one of the several factors which stimulate the secretion of growth and nerve growth factors in patients with Multiple Sclerosis (MS). Therefore, the aim of this study was to evaluate the effect of 12 weeks of Pilates training and ginger consumption on serum level of BDNF and TNF-α in women with MS.
MethodsForty women with MS were randomly selected from 85 women referred to the Welfare Office of Urmia city, in Iran, with the age range of 30-35 in the form of a pre-test and post-test quasi-experimental study design. Participants were divided into four groups including: exercise+ supplement, exercise+ placebo, ginger supplement and control group with 10 individuals in each group. The intervention groups performed Pilates exercises for 12 weeks, 3 sessions of 60 minutes every week, and the supplement groups took three ginger capsules of one gram daily. In order to analyze the data, analysis of covariance was done using SPSS-22 software.
ResultsThe results showed that ginger consumption combined with physical exercises increases BDNF in the intervention groups compared to the control groups, but this increase is more significant in the exercise+supplement group (p<0.05) than other groups. Also, the results showed that TNF-α level has a significant decrease in the intervention group compared to the control group (p<0.05).
ConclusionThe results showed that ginger consumption as a non-invasive method can have a positive effect on increasing BDNF level and decreasing TNF-α level in women with MS.

Osteoarthritis as a result of the destruction of articular cartilage, leads to joint pain and stiffness. The purpose of this study was to investigate the effect of endurance and ozone therapy on the expression levels of FLS and VEGF genes in the knee tissue of rats with knee osteoarthritis.
MethodsSubjects were male Wistar rats that were randomly divided into 5 groups after two weeks of adaptation, including: control-healthy, control-patient, patient-ozone, patient-exercise and patient-sport. For adapting the rats to exercise and treadmill, walking program was performed before exercise program. The training program consisted of 30 minutes of running on a treadmill with no slope at a speed of 16 m/min for the first week and one m/min added weekly until it reached 22 m/min in the eighth week. Ozone was injected into the knee through the tibiofemoral articular line at a concentration of 20 μg/ml once a week for 3 weeks at 9 ochr('39')clock in the morning. Expression levels of FLS and VEGF genes were measured by Real Time PCR. One way ANOVA and Tukeychr('39')s method were used to determine the difference between groups.
ResultsThe results showed that the expression levels of FLS and VEGF genes were significantly increased in rats with osteoarthritis. It was also shown that physical training and ozone therapy led to a decrease in FLS and VEGF gene expression levels in osteoarthritic rats. VEGF and its receptors are expressed in OA cartilage and indicating the possibility that VEGF is involved in the elimination of OA articular cartilage by increasing MMP production.
ConclusionThe results of this study are promising for the role of VEGF as a diagnostic marker. VEGF can participate in early OA changes and a treatment modality by modulating VEGF production, which can be considered in future.

Foxo1 is a transcription factor in insulin signaling in the liver. The function of Foxo1 is important in the ability of insulin to regulate hepatic glucose production and glucose homeostasis. The aim of this study was to determine the effect of resistance training program on Foxo1 expression in liver cells as well as glucose and insulin serum levels in male Wistar rats with type 2 diabetes.
MethodsThe statistical population of this experimental study was 10-weeks old, male Wistar rats weighing 220±20 g (n=14). The studied rats were subjected to 6 weeks of high-fat diet and STZ injection for type 2 diabetes induction and then rats were randomly divided into two groups (control and resistance). The training group participated in a resistance training course for 6 weeks, 5 sessions per week, in the form of 4 sets and 5 repetitions per set, climbing a 26-step ladder to a height of one meter with a vertical slope of 80%. Finally, 48 hours after the last training session, fasting blood glucose and insulin levels as well as Foxo1 expression level in liver cells of both groups were measured and compared by independent t-test (a=%5).
ResultsThe resistance training improved fasting blood glucose compared to control subjects (p>0.001). the serum insulin levels were significantly increased (p=0.042) and Foxo1 expression significantly decreased in resistance group when compared with control subjects (p=0.023).
ConclusionBased on these data, the decrease in glucose concentration in the exercise group can probably be attributed to the decreased expression of Foxo1 in hepatocyte cells in response to resistance training.

Mitochondrial dysfunction is one of the main risk factors for neurological diseases which are associated with aging. On the other hand, aerobic exercise has beneficial effects on the brain health and cognitive function, and also improves mitochondrial dynamics. Therefore, the aim of the present study was to investigate the effect of 4 weeks of aerobic exercise on spatial learning, memory performance and mitochondrial dynamics in the hippocampal tissue of old rats.
MethodsFor this purpose, 14 male Wistar rats at 20 months of age were randomly divided into 2 groups: aerobic exercise (n=7) and control group (n=7). The exercise group performed 4 weeks of treadmill training (5 days per week at a speed of 10 to 15 m/min). Forty-eight hours after the last training session, the animals underwent behavioral tests. Twenty-four hours after the behavioral test, all rats were killed and hippocampal tissue was extracted. The mRNA expression of OPA1, Mfn2 and Drp1 genes were assayed using Real Time-PCR. The Independent t test was used for statistical analysis.
ConclusionAerobic exercise in old animals improved spatial learning and memory performance, increased hippocampal OPA1 gene expression, and decreased Drp1 gene expression compared to the control group (p≤0.01).
ConclusionIt seems that aerobic exercise can improve the function of brain mitochondria by modulating fusion and fission processes and it can be considered as an effective non-pharmacological method to deal with aging-related learning and memory perturbations.

The liver is one of the most important organs in the body and plays an essential role in the metabolism of drugs and toxins. Therefore, any damage to the liver can disrupt its function and cause various diseases. On the other hand, due to the increasing use of the medicinal herbs, in this study, the effects of hepatic protection of aqueous extracts of Humulus lupulus on liver damage induced by carbon tetrachloride in rats were investigated.
MethodsIn this experimental study, 36 male Wistar rats were divided into 6 groups (n=6). Group 1 (normal control) and Group 2 (control CCl4): administered with distilled water orally for 14 days. Groups 3, 4, and 5 received 50, 100, and 200 mg/kg of Humulus lupulus extract for 14 days respectively. Group 6: administered with silymarin (as standard drug) 100 mg/kg orally for 14 days. All of the groups except group 1 received 1 ml CCl4 (1:1 v/v dissolved in olive oil) on 14th day. All animals were sacrificed and blood samples were collected to determine the biochemical parameters.
ResultsPretreatment with a Humulus lupulus extract significantly reduced the serum levels of ALT, AST, ALP, TG, and increased the serum HDL levels, but did not have a significant effect on GGT and LDL levels.
ConclusionThe results of this study showed that the Humulus lupulus extract can protect the liver from damage caused by carbon tetrachloride and have beneficial effects on lipid profile.

Based on the probably positive role of exercise on total oxidative status (TOS) and total antioxidant system (TAS) and their effect on the process of angiogenesis, the aim of the current study was to evaluate the effect of one course of moderate-intensity endurance training on the gene expression level of vascular endothelial growth factor-B (VEGF-B) and angiopoietin-1(ANGPT-1) and TAS and TOS status in cardiac tissue of male rats.
MethodsIn this study, 20 male Wistar rats were divided into two equal groups as follows: 1-training group and 2- control group: no sports activities were performed on them. Rats in the training group performed moderate endurance training for 6 weeks and 5 sessions per week from the twelfth week of life. Twenty-four hours after the last training session, heart tissue samples were extracted to measure gene expression levels of VEGF-B and ANGPT-1 and the TAS and TOS status in heart tissues. T-test with the statistical level of (p˂0.05) was used for between groups comparison.
ResultsFindings showed that after six weeks of endurance training, the expression level of ANGPT-1 and VEGF-B and the amount of TAS in the exercise group significantly increased (p=0.001) and the amount of TOS significantly decreased compared to the control group (p=0.008).
ConclusionAccording to the results of the present study, it seems that moderate-intensity endurance training can be effective in preventing cardiovascular disease by increasing factors involving in angiogenesis, improving TAS and reducing TOS in heart tissue of rats.

Major advantage of the technique relies on a nominated drug molecule that is applied for pharmacodynamics optimization due to lack of serious pharmacokinetics challenges. According to the importance of the subject, a present contribution has been dedicated to the in-silico analysis of a few drug classes with the aim of achieving potential anti-leishmanial pharmacophores.
Methods3D structure of protein targets within leishmania parasite were retrieved from Brookhaven Protein Data Bank (PDB) on the basis of literature reports to evaluate the related complexes with drugs via molecular docking. Qualitative and quantitative analysis of drug-target interaction patterns in docked complexes offered drugs with higher binding affinities toward targets and finally structural patterns or hypothetical anti-leishmanial pharmacophores were proposed with regard to the top-ranked pharmaceutical compounds.
ResultsHighest free binding energy could be estimated for Nateglinide in binding to farnesyl diphosphate synthase (ΔGb -13.30 kcal/mol). Among steroids, Norgestrel synthase (ΔGb -9.48 kcal/mol) and Testosterone synthase (ΔGb -8.05 kcal/mol) exhibited higher enzyme binding affinities and Arg82 was a key residue in making hydrogen bonds. Within fused tricyclic structural patterns, mirtazapine exhibited highest binding affinity to deoxy uridine triphosphate (ΔGb -8.64 kcal/mol). In Carbamazepine, amide substituent of the central ring facilitated the formation of two effective hydrogen bonds with Gln21 and Asn25 in deoxy uridine triphosphate.
ConclusionOn the basis of obtained results for steroids and fused tricyclic scaffolds, it will be possible to design molecules that can inhibit several pathogenic targets simultaneously.

ABCA1 plays an important role in HDL biosynthesis and cellular cholesterol homeostasis and is effective in preventing the formation of atherosclerotic plaques. This study aimed to investigate the effect of combined and rehabilitation training on ABCA1 gene expression in blood lymphocytes and lipid profile in middle-aged men with coronary bypass graft.
MethodsIn this quasi-experimental study, 45 middle-aged men (mean age: 55.58±4.7 years) were selected by convenience sampling method and randomly divided into three groups of combined training (15 people), rehabilitation (n=15), and control (n=15). The combined training group included resistance and aerobic exercises and rehabilitation training group included aerobic exercises for 8 weeks (3 sessions per week). Blood samples were taken from all subjects at weeks 0 and 8 to measure the expression levels of ABCA1, HDL, and LDL genes. Kruskal-Wallis, one-way ANOVA, and Tukey post hoc tests were used for statistical analysis using SPSS software, and significance level was p≤0.05 .
ResultsThe results showed that eight weeks of combined training and rehabilitation training increased the expression level of the ABCA1 (p=0.001 and p=0.001, respectively) gene and plasma HDL (p=0.001 and p=0.012, respectively) level and decreased plasma LDL (p=0.001 and p=0.01, respectively) level. There was also a significant difference between the three groups in ABCA1 and HDL values in favor of the combined training group.
ConclusionIt seems that combined training can be more effective than rehabilitation training to further increase ABCA1 gene expression and plasma HDL levels and decrease LDL level in middle-aged men with coronary bypass graft.

Breast cancer is one of the most common diseases and the leading cause of death among women worldwide. The aim of the present study was to determine the effects of five weeks of endurance training in combination with curcumin on the cancer progression, intratumoral gene expression of angiomiR-126 and Angiopoietin-1 in breast cancer bearing female BALB/c mice.
MethodsThe present study was an experimental study. 4T1 breast cancer was transplanted into forty female BALB/c mice. Afterward, they were randomly divided into four groups including, a control group (C), an endurance training group (E), an endurance training along with curcumin (EC) and curcumin group (CC). E and EC groups performed 40 min at a constant running intensity corresponding to 60-65% vVo2peak at 0% inclination, 5 weeks (five days a week). CC and EC groups were treated by oral gavage with curcumin for five weeks (six days a week). Then, tumor tissue of all mice was extracted 24 hour after the last training session. Gene expression levels of miR-126 and angiopoietin-1 were determined by qReal-time PCR. Statistical data values were also measured by One-way ANOVA.
ResultsThe results showed a significant inhibition of cancer growth, increased gene expression of miR-126 (p<0.001) and decreased angiopoietin-1 gene expression (p<0.001) in E, CC, and especially E-C compared to control group. All of these changes were significantly greater in the EC group than in the other intervention groups (p<0.001).
ConclusionIt seems that five weeks of endurance training with curcumin supplementation possibly have a further effect on decreasing breast cancer mass growth by inhibiting miR-126 /angiopoietin-1 axis compared to other interventions alone.

Over the last couple of decades, research has focused on attempting to understand the genetic influence on sports performance. Whole exome sequencing (WES) has great potential to explore all possible genetic variants associated with physical performance. However, this new technique has not been used in sports genetics research. Therefore the aim of this case study was to report new variants in genes involved in skeletal muscles strength (ACE, NOS3, IGF1R, IL-6) in an elite weightlifter using WES.
MethodsAfter obtaining a written testimony from a 32-year-old man, who is a member of the Iranian national weightlifting team, 5 cc of blood was taken and then DNA was extracted. DNA samples were analyzed using the whole exome sequencing method.
ResultsWhole exome sequencing analysis revealed a missense variant (99454613G>A) in IGF1R, 22771156C>T polymorphism in IL-6, 150695726T>C and 150704250C>G polymorphisms in NOS3 and 61573761T>C and 61564052A>G in ACE genes.
ConclusionResults of whole exome sequencing analysis revealed a missense variant and polymorphism in muscle strength and hypertrophy related genes in this elite weightlifter. However, other studies with larger sample sizes are needed to investigate the relationship between these variants and power athlete status.
