Evaluation of antinociceptive effects of a selective iNOS inhibitor and imipramine in sciatic nerveligated mice by hot-plate test

Abstract:
Introduction
The underlying mechanisms of neuropathic pain are poorly understood. Because selective inducible nitric oxide synthase inhibitor (Aminoguanidine) alone and on combination with imipramine on sciatic nerve- ligated mice by hotplate test.
Materials And Methods
This work was accomplished on sixty four (64) male mice that were divided randomly in aminoguanidine، imipramine، saline، aminoguanidine-imipramine received groups (eight mice in each). All drugs were dissolved in saline and were injected intraperitoneally (i. p). Pain sensitivity was assessed on day 14 after sciatic nerve ligation، using the hot-plate test before and following drug administrations for two hours.
Results
were observed following the sciatic nerve ligation، 14 days after surgery. Administration of aminoguanidine (50 and 100 mg/kg، i. p) animals. Latency periods after co-administrations of aminoguanidine and imipramine increased animals.
Conclusion
that inducible nitric oxide synthase may play an important role in the development of such sciatic nerve ligated-evoked pathological pain conditions. However، further studies with more underlying mechanisms of neuropathic pain are recommended.
