Agricultural Waste using for Cellulase enzyme production to increase by the fungus Trichoderma reesei

Background And Objective
Concern to the importance and application of cellulase enzymes, finding the best condition for maximum enzyme production with low cost is one the most important step in enzyme production. In this research, the production of cellulase enzymes by Trichoderma reesei on cheap lignocellulosic substrates (corn Crust), from agricultural waste, was studied using solid state fermentation.
Material And Methods
The fungus Trichoderma reesei, microorganisms, used in this study, were obtained from collection of fungi and bacteria, Iranian Research Organization for Science and Technology (IROST, accession number: PTCC 5142) in lyophilized ampoules. Agricultural waste for the production of the enzyme was used as the solid substrate. 10 g of solid substrates were moistened with 30 ml of saline solution (in g) was wet. The plates containing the Trichoderma inoculum was transferred to a sterile solid medium, and then incubated. Extraction of cellulase enzymes produced by solid substrate buffer solutions was made. FP activity and CMCase enzymes activity were measured. Also enzyme biochemical properties were investigated.
Results
It was seen that Fpase enzyme production was more than CMCase. Two enzyme productions in salt solution A were much more than salt solution B and C.
Conclusion
The results showed that the enzyme produced by T. reesei (PTCC 5142) in optimal conditions during 7 days at 32 °C in corn crust and A salt solution was 12098.4 u/gds. Statistical analyzes showed that the best conditions in terms of stability of cellulase enzymes is at 28 °C
